肌球蛋白轻链激酶mRNA在非小细胞肺癌患者外周血的表达情况及其临床意义
Expression and clinical significance of myosin light chain kinase message RNA in peripheral blood of patients with non-small cell lung cancer
【摘要】 目的探讨肌球蛋白轻链激酶(MYLK)mRNA在非小细胞肺癌(NSCLC)患者外周血的表达情况及其与临床特征的关系。方法 48例NSCLC患者(肺癌组)和48例非肿瘤患者(对照组),抽取静脉血后分离出单个核细胞,使用实时荧光定量PCR检测MYLK mRNA的表达情况,并分析其表达量与临床特征的关系。结果肺癌组及对照组患者外周血均有MYLK mRNA表达,肺癌组患者MYLK mRNA表达量为(1.40±0.57),明显低于对照组的(2.03±0.16)(P<0.05);术前组与术后组、鳞癌组与腺癌组、高+中分化组与低分化组、男性组与女性组、吸烟组与不吸烟组的MYLK mRNA相对表达量比较,差异均无统计学意义(P均>0.05);Ⅲ+Ⅳ期组患者、有淋巴结转移组患者MYLK mRNA相对表达量分别高于Ⅰ+Ⅱ期组、无淋巴结转移组患者(P<0.05)。结论外周血MYLK mRNA的表达水平可能与NSCLC发生发展有关,对判断NSCLC预后有一定价值。
【Abstract】 Objective To explore the expression of myosin light chain kinase( MYLK) message RNA( mRNA) in peripheral blood of patients with non-small cell lung cancer( NSCLC),and to investigate the relationship of MYLK mRNA expression with clinical characteristics. Methods Mononuclear cells were isolated from the venous blood of 48 patients with NSCLC( lung cancer group) and 48 patients without tumor( control group). And then the expression of MYLK mRNA was detected by real-time quantitative PCR,and the relationship between the expression level of MYLK mRNA and clinical characteristics was analyzed. Results MYLK mRNA was expressed in peripheral blood of the patients in lung cancer group and control group. The expression level of MYLK mRNA of the patients in lung cancer group was( 1. 40 ± 0. 57),and was significantly lower than that of control group( 2. 03 ± 0. 16)( P < 0. 05). For patients with NSCLC,there was no significant difference in the relative expression level of MYLK mRNA between two different subgroups,including preoperative group and postoperative group,squamous cell carcinoma group and adenocarcinoma group,well-differentiated + moderated differentiated group and poor-differentiated group,male group and female group,smoking group and non-smoking group( P > 0. 05). The relative expression levels of MYLK mRNA in patients with stage Ⅲ + Ⅳ and with lymphatic metastasis were significantly higher than those in patients with stageⅠ+Ⅱ and without lymphatic metastasis respectively( P < 0. 05). Conclusion The expression level of MYLK mRNA in peripheral blood may be closely related to the occurrence and development of NSCLC,and it is valuable for evaluating the prognosis of NSCLC to a certain extent.
