肿节风对放射性肺损伤小型猪氧化应激反应的干预作用研究

【摘要】 目的:探讨肿节风对放射性肺损伤小型猪氧化应激反应的干预作用。方法:将75头小型猪按随机数字表法分为空白组、单照组及药照组3组,每组25头。药照组于照射前1周开始持续给予肿节风配方颗粒直至取出右肺,其余两组给予等量生理盐水;在麻醉条件下单照组和药照组给予单次15Gyγ射线照射右肺,空白组只麻醉不予照射;3组分别于照射后2周、4周、8周、12周、24周随机取5头处死并取右肺,称重后制成组织匀浆,再测定组织匀浆中活性氧簇(ROS)、丙二醛(MDA)含量及总超氧化物歧化酶(T-SOD)活性。结果:单照组和药照组ROS含量及T-SOD活性均先上升后下降,而MDA含量则一直上升;分别与空白组比较时,单照组ROS及MDA含量分别从第2周和第4周起差异有统计学意义(P<0.05),T-SOD活性则在第4周、第12周和第24周差异有统计学意义(P<0.05),而药照组ROS及MDA含量分别在第8周前和第12周后差异有统计学意义(P<0.05),T-SOD活性则只在第24周时差异无统计学意义(P>0.05);与单照组比较,药照组ROS含量、TSOD活性及MDA含量分别从第2周、第4周和第8周起差异有统计学意义(P<0.05)。结论:在放射性肺损伤的发病过程中,肿节风通过清除ROS、抑制MDA生成和上调T-SOD活性,减轻氧化应激反应,从而发挥放射防护作用。

【Abstract】 Objective:To investigate the effect of Sarcandra glabra on oxidative stress in miniature pig with radiation-induced lung injury.Methods:75miniature pigs were divided into the control group,the radiation group and the radiation plus medication group.Miniature pigs in radiation plus medication group were treated with Sarcandra glabra from 1week before exposure of the right lung to 15Gyγ-ray irradiation,while those in the other two groups received an equal amount of saline.Right lung were taken in the weeks 2,4,8,12 and 24after the irradiation,and reactive oxygen species(ROS),malondialdehyde(MDA)and total superoxide dismutase(T-SOD)were measured in lung tissue.Results:ROS content and T-SOD activity were increased at earlier time points and then decreased at later time points,while MDA content was increased persistently both in the radiation group and the radiation plus medication group(P <0.05).Compared with control group,the contents of ROS and MDA showed significant difference from the 2nd week and 4th week,and T-SOD activity showed significant difference in the 4th week,12 th week and 24 th week in the radiation group,respectively(P <0.05).While in radiation plus medication group,T-SOD activity only showed significant difference at the 24 th week(P <0.05),and ROS and MDA content showed significant difference before the 8th week and after 12 th week,respectively(P <0.05).Compared with the radiation group,the activities of ROS,MDA and T-SOD showed significant difference from the 2nd week,4th week and 8th week in the radiation plus medication group, respectively(P < 0.05).Conclusion: Sarcandra glabra alleviated oxidative stress and played a protective role in radiation-induced lung injury in miniature pig by scavenging reactive oxygen species,restraining the production of MDA and enhancing the activity of T-SOD.
