碘化物过量对人神经细胞的损伤作用及机制研究
Excess Iodide Effects on Autophagy of Human Nerve Cells(SH-SY5Y)
【摘要】 目的探讨碘化物过量对人神经母细胞瘤细胞(SH-SY5Y)的损伤作用及其机制。方法将体外培养的SH-SY5Y细胞用0(对照)、10、20、30 mmol/L碘化钾(KI)分别染毒处理24 h后,采用CCK-8法、单丹黄酰尸胺(MDC)染色法及Wsetern blot实验检测细胞存活率、自噬囊泡生成以及自噬相关蛋白表达情况。结果与对照组相比较,30 mmol/L碘化物染毒组细胞存活率明显下降(P<0.05),自噬囊泡生成明显增多(P<0.05),自噬相关蛋白Beclin1表达明显升高(P<0.05)。结论一定剂量的碘可降低神经细胞的存活率,诱导细胞自噬,碘化物过量对神经细胞的损伤作用可能与其诱导活化细胞自噬有关。
【Abstract】 Objectives To investigate the damage of excess iodide on human neuroblastoma cells( SH-SY5Y)and its related mechanism. Methods SH-SY5 Y cells were treated with potassium iodide( KI) at the concentrations of 0,10,20 and 30 mmol / L in the medium for 24 hours. The viability of cells,the generation of autophagic vesicles and the expression of Beclin1 were tested by CCK- 8 assay,monodansylcadaverine( MDC)- staining assay and western blot assay. Results Compared with the control group,the viability of cells in the 30 mmol / L KI group was decreased obviously( P < 0. 05),the generation of autophagic vesicles was increased( P < 0. 05) and the expression of Beclin1 protein was higher significantly( P < 0. 05). Conclusions Excess iodide can notablely decrease the viability of SH-SY5 Y cells,increase the generation of autophagic vesicles and increase the expression of Beclin1 protein in SH-SY5 Y cells. These results suggested that the damage of nerve cell induced by excess iodide might be related to autophagy.
