芦竹组培快繁研究初探

【摘要】 以芦竹的幼叶、幼茎和嫩叶鞘为外植体,研究了外植体类型、外源激素对芦竹愈伤组织诱导、分化、芽体增殖的影响。结果表明:愈伤组织诱导的外植体以幼叶较好;愈伤组织诱导培养基以MS+1.5mg/L 2,4-D+0.5mg/L NAA+0.05mg/L KT为最适,出愈率高达(92.22±1.11)%;愈伤组织分化培养基以MS+0.2mg/LNAA+2.0mg/L6-BA为宜,芽的分化率达到40.00%;芽体增殖培养基以MS+4.0mg/L6-BA为宜,增殖系数达到(3.10±0.02);再生植株移栽到泥炭∶田园土=1∶1的基质中成活率达到95%以上。

【Abstract】 Explants of young leaf, stem and leaf sheath of Arundo donax, comparing the influence of callus induction and differentiation with different explant type and exogenous hormone. The results showed that(1) Young leafs was suitable for explant;(2) The best callus-inducing medium was MS+1.5mg/L 2,4-D+0.5 mg/L NAA+0.05mg/L KT,and the callus rate was(92.22±1.11) %;(3) MS+0.2mg/L NAA+2.0 mg/L 6-BA were used for the best callusdifferentiation medium in turn the bud differentiation stage reached 40.00%;( 4) As for the buds multiplication medium, MS +4.0 mg/L6-BA were appropriate, and multiplication coefficient was up to the(3.10 ±0.02) on this condition;( 5) When transplant the aseptic seedling to a peat: garden soil = 1:1 matrix, the survival rate could be reach up to 95%.
