甘草甜素对人乳腺癌细胞MDA-MB-231生长的影响
Effects of glycyrrhizin on the growth of human breast cancer cells of MDA-MB-231
【摘要】 目的研究甘草甜素对人乳腺癌细胞MDA-MB-231生长的影响。方法将对数生长期人乳腺癌细胞随机分为5组:对照组仅加等量培养液,实验A组、B组、C组及D组分别给予浓度分别为0,50,100,200μmol·L-1的甘草甜素细胞培养液;用噻唑蓝(MTT)法检测5组人乳腺癌细胞MDA-MB-231增殖、凋亡情况,并采用半定量逆转录-聚合酶联反应(RT-PCR)检测细胞中Fas、survivin、Bcl-2、Bax和caspase-3基因水平。结果随着甘草甜素浓度的增加和作用时间的延长,对人乳腺癌细胞MDA-MB-231增殖抑制作用显著增强,其中48 h后对照组OD值为(0.61±0.21),实验A、B、C、D组分别为(0.61±0.11),(0.56±0.15),(0.48±0.17),(0.45±0.14),实验A、B、C、D组的抑制率分别为8.17%,10.01%,24.41%,31.82%,各组比较差异有统计学意义(P<0.05);Fas、survivin、Bax和caspase-3基因表达因甘草甜素浓度增加而降低,而Bcl-2表达则因甘草甜素浓度增加而上升。实验D组的Fas、survivin、Bax和caspase-3基因表达下降程度明显高于其他组(P<0.05),Bcl-2基因表达水平增加程度也明显高于其他组(P<0.05)。结论甘草甜素对人乳腺癌细胞MDA-MB-231细胞增殖抑制作用强,具有明显的量效和时效关系。
【Abstract】 Objective To explore the effects of glycyrrhizin on the growth of human breast cancer cells of MDA- MB- 231. Methods The human breast cancer cells in the logarithmic phase of growth were divided into 5 groups at random,among which the control group were only given the same amount of culture solution,and the test A,B,C and D group were given glycyrrhizin cell culture medium respectively,of which the concentrations were respectively 0,50,100 and 200 μmol · L-1. The proliferation and apoptosis of human breast cancer cell line MDA- MB-231 in the 5 groups were observed by the tetrazolium( MTT) assay. The levels of Fas,survivin,Bcl- 2,Bax and caspase- 3 were detected by semi quantitative reverse transcription polymerase chain reaction( RT- PCR). Results With the increase of the concentration of glycyrrhizin and the extension of the action time,the proliferation inhibition of human breast cancer cells of MDA- MB- 231 were significantly enhanced,and the OD value in the control group was( 0. 61 ± 0. 21)after 48 h,of which the test A group was( 0. 61 ± 0. 11),with the test Bwas( 0. 56 ± 0. 15),the test C was( 0. 48 ± 0. 17),the test D was( 0. 45 ± 0. 14). The inhibition rate in the test A group was 8. 17%,with the test B group was 10. 01%,the test C group was 24. 41%,the test D group was 31. 82%,and the differences in each two groups were statistically significant( P < 0. 05). The gene expre-ssions of Fas,survivin,Bax and caspase- 3 were decreased with the increase of the concentration of glycyrrhizin,among which the gene expressions of Fas,survivin,Bax and caspase- 3 in the test D group were significantly higher than other groups( P < 0. 05).While the incerased level of the expression of Bcl- 2 was significantly higher than the other groups( P < 0. 05). Conclusion The inhibitory effects of MDA- MB- 231 on the proliferation of human breast cancer cell line are strong,which has obvious dose and time- response relationship,and the mechanism of action is significantly correlated with the Survivin,Bax,Caspase- 3,Fas,Bcl- 2 and other genes of apoptotic cells.
