黑龙江省2012-2015年诺如病毒感染的分子流行病学研究

【摘要】 目的监测2012-2015年黑龙江省诺如病毒腹泻的流行情况及流行株的基因型、重组变异特征。方法 2012年1月-2015年12月共采集480份哨点医院腹泻病人的粪便标本,采用Real-time PCR检测诺如病毒GI和GII基因组,对GII基因组阳性标本经传统RT-PCR扩增诺如病毒部分RNA依赖的RNA聚合酶(Rd Rp)和衣壳蛋白(VP1)序列,对核苷酸序列进行基因分型。结果 2012-2015年采集的腹泻病人粪便标本诺如病毒阳性率为3.54%(17/480),GI阳性1株,GII阳性16株。其中11株GII阳性株扩增测序成功,分为6种基因型,分别为:GII.4 Sydney2012型、GII.4 Den Haag2006b型、GII.P7/GII.6型、GII.P12/GII.3型、GII.P16/GII.13型和GII.17型。结论黑龙江省2012-2015年诺如病毒存在多种基因型,并存在不同基因型间重组株,提示诺如病毒在黑龙江省的遗传变异日趋复杂。

【Abstract】 Objective To monitor the epidemiology of norovirus in patients with diarrhea in Heilongjiang province from2012 to 2015, and to characterize the genotypes of norovirus strains. Methods 480 fecal specimens of viral diarrhea cases at sentinel hospitals between January 2012 and December 2015 were collected.Real-time PCR was used to screen for norovirus genogroups GI and GII. The genotypes of norovirus genogroup GII were classified based on the nucleotide sequences of both partial polymerase and capsid fragments. Results Among 480 fecal specimens,17 strains(3.54%)were tested positive for norovirus according to real-time PCR,1 strain was positive for norovirus GI,and 16 strains were positive for norovirus GII.Six distinct genotypes were identified in 11 norovirus GII strains, including GII.4 Sydney2012、GII.4Den Haag2006b、GII.P7/GII.6、GII.P12/GII.3、GII.P16/GII.13 and GII.17. Conclusion The circulating norovirus showed genetic diversity.A variety of recombinant viral strains were detected,suggesting that the complexity of the genetic variation of norovirus were increasing in Heilongjiang province.
