磁珠富集法开发长臀鮠微卫星分子标记

【摘要】 为长臀鮠遗传育种、种质鉴定和种苗流放等提供理论依据,采用磁珠富集法开发长臀鮠(Cranoglanis bouderius)微卫星分子标记,筛选获得阳性克隆进行分析,选取侧翼较长的序列,用Primer Premier 5.0设计合成引物。结果表明:从596个白斑中筛选获得80个阳性克隆,测序获得微卫星序列47个,其中完美型31个(66%),非完美型14个(30%),复合型2个(4%)。设计合成32对引物,通过优化反应条件,得到27对引物能扩增特异条带,其中多态性引物24对。

【Abstract】 To provide the theoretical basis for genetic breeding,germplasm identification and releasing.Magnetic beads enriched method was used to isolate microsatellites fromC.bouderius,through sequencing and analysis the positive colonies,the repeats which contained enough flanking region were obtained to design 32 microsatellite primers by the software Primer Premier 5.0.Results:80positive colonies were obtained through 596 white spots.Sequencing of the 80 positive colonies confirmed that 47 microsatellite sequences were obtained.From these sequences,31sequences(66%)were perfect repeats,14(30%) were imperfect repeats,and two sequences(4%)were compound repeats.27 pairs of microsatellite primers used successfully to amplify special fragments at appropriate annealing temperature,and 24 pairs of them were polymorphism within species.
