子宫内膜异位症与线粒体能量代谢关系的研究

【摘要】 目的探究子宫内膜异位症(内异症)的发生发展与线粒体能量代谢的关系。方法收集20例内异症患者的在位内膜、盆腔子宫内膜异位灶和卵巢子宫内膜异位囊肿壁组织为异位症组,20例非内异症患者的在位内膜作为对照组,根据差速离心法提取线粒体DNA,采用实时荧光定量PCR扩增方法来检测线粒体DNA中细胞色素B(Cyt B)和烟酰胺腺嘌呤二核苷二钠(NADH)的表达,并比较不同分期、年龄及疼痛程度对Cyt B和NADH表达的影响。结果异位症组患者异位内膜Cyt B和NADH的表达高于对照组、异位症在位内膜和异位囊肿组(P<0.05),内异症Ⅰ、Ⅱ期和Ⅲ、Ⅳ期患者异位内膜的Cyt B和NADH表达高于对照组(P<0.05),不同年龄段异位内膜患者CytB和NADH的表达均高于对照组(P<0.05)。不同疼痛程度患者异位内膜CytB和NADH的表达均高于对照组(P<0.05)。结论线粒体能量代谢可能与内异症的发生发展相关,为内异症的演变提供了生物能量。

【Abstract】 Objective To investigate the relationship between the edometriosis and mitochondrial energy metabolism.Method Collecting eutopic endometrium,ectopic endometrium in abdominal sites,ovarian cyst tissue of EMs patients(EMs group)and normal endometrium from patients without EMs(control group).The expression of mitochondrial cytochrome B(Cyt B)and dihydronicotinamide-adenine dinucleotide disodium salt(NADH)was detected by using fluorescence quantitative PCR amplification method to the relationship between Cyt B,NADH and clinical characteristic like stages,age,pain were analyzed.Result The expression of Cyt B and NADH of ectopic endometrium was higher than control group,eutopic endometrium and ovarian cyst(P<0.05).The expression of Cyt B,NADH in ectopic endometrium in the stageⅠ、Ⅱand Ⅲ、Ⅳ was higher than control group(P<0.05).The expression of Cyt B and NADH in ectopic endometrium was significantly higher than control group in different ages(P<0.05).The expression of Cyt B and NADH in ectopic endometrium was higher than control group which was independent on the pain degree(P<0.05).Conclusion The mitochondrial activity may provide energy for endometriosis thus enhance the development of endometriosis.
