长链非编码RNA CHL1-AS2在卵巢子宫内膜异位症中的表达及意义初探

【摘要】 目的检测长链非编码RNA(lncRNA)CHL1-AS2分别在卵巢子宫内膜异位症(EMs)患者的卵巢内异症病灶(EC)、病灶旁组织(NO)及配对在位内膜(EU)的表达,以期对卵巢EMs的发病机制进行一定的探讨。方法收集10例卵巢EMs患者以上三种组织,另外15例患者的卵巢内异症病灶及病灶旁组织,通过实时荧光定量PCR(qPCR)检测CHL1-AS2的表达量并比较各组织间表达差异。结果 lncRNA CHL1-AS2在卵巢EMs患者的在位内膜低表达,卵巢内异症病灶及病灶旁组织高表达,EC/EU与NO/EU的表达倍比差异无统计学意义,且CHL1-AS2在卵巢内异症病灶和病灶旁组织的表达量趋近,同时月经周期不影响CHL1-AS2的表达倍比。结论 lncRNA CHL1-AS2在卵巢EMs的内异症病灶、病灶旁组织及在位内膜的表达有差异,可能与卵巢EMs的发病机制有关。

【Abstract】 Objective To explore the expression of long non-coding RNA(lncRNA)CHL1-AS2 in ovarian ectopic endometrial tissue,adjacent tissue and paired eutopic endometrial tissue of endometriosis patients.Methods The above three kinds of tissues in ten patients with ovarian endometriosis,ectopic endometrial tissue and adjacent tissue in another fifteenth patients were collected.Real-time fluorescent quantitative PCR(qPCR)was used to examine the expression ofCHL1-AS2,and the expression differences in each tissue was compared.Results CHL1-AS2 was high expressed in ectopic endometrium and adjacent tissue but low expressed in eutopic endometrium.It showed no statistical significance when compared the value of EC/EU and NO/EU.And it revealed a tendency that the expression level of CHL1-AS2 in ectopic endometrium and adjacent tissue were nearly the same.Besides,menstrual cycle had no effect on the expression of CHL1-AS2 in the three kinds of tissues.Conclusions The study showed that there was difference in the expression of the long non-coding RNA,CHL1-AS2,in ovarian ectopic endometrial tissue,adjacent tissue and paired eutopic endometrial tissue.CHL1-AS2 may contribute to the pathogenesis of ovarian endometriosis.
