光敏剂五聚赖氨酸-β-羰基酞菁锌对宫颈癌Hela细胞的光动力学治疗研究

【摘要】 目的研究光敏剂五聚赖氨酸-β-羰基酞菁锌"[ZnPc-(Lys)5]光动力诱导宫颈癌Hela细胞死亡的方式。方法测量Hela细胞对光敏剂ZnPc-(Lys)5的摄取、光毒性和暗毒性,利用流式细胞仪分析光动力疗法对Hela细胞的影响,包括其细胞凋亡和细胞坏死的比例;并通过透射电镜观察细胞超微结构的改变情况。结果 Hela细胞对该光敏剂的摄取随着ZnPc-(Lys)5浓度的升高而增强,相关光动力学实验结果显示:光敏剂ZnPc-(Lys)5对宫颈癌Hela细胞的光动力疗效作用理想,无暗毒性。其介导的光动力治疗是通过诱导细胞凋亡途径而达到消灭肿瘤细胞的作用。ZnPc-(Lys)5对Hela细胞光动力作用后随作用的不同时间,其凋亡的比率有显著性差异:0.5h的早期凋亡率为(50.36±1.02)%,1h为(40.80±0.72)%,2h为(30.87±2.80)%,4h为(26.87±0.81)%,各组比较差异均有统计学意义(P<0.05)。晚期凋亡率0.5h为(34.23±0.55)%,1h为(33.97±1.40)%,2h为(45.57±1.12)%,4h为(45.57±1.22)%,第0.5h、1h与第2h、4h比较,差异有统计学意义(P<0.05)。Hela细胞在光动力作用下1h超微结构主要表现为早期凋亡,2h后超微结构改变为晚期凋亡和胀亡的特征改变。结论光敏剂ZnPc-(Lys)5介导的光动力治疗主要通过凋亡途径特异性诱导宫颈癌Hela细胞的死亡。

【Abstract】 Objective To investigate the mechanism of the tumor-targeting photosensitizerβ-carbonylphthalocyanine zinc ZnPc-(Lys)5on Hela cells in photodynamic therapy.Methods Measured the cellular uptakes of the photosensitizerβ-carbonylphthalocyanine zinc ZnPc-(Lys)5into Hela cells,as well as the phototoxicity and the dark toxicity on Hela cells.In addition,the proportion of cell apoptosis and necrosis were investigated by flow cytometry and transmission electron microscope.ResultsThe cellular uptakes of the ZnPc-(Lys)5into Hela cells were increased with the increase of the concentration of photosensitizer.This was consistent with the results in its photodynamic therapy studies.In photodynamic therapy,the mechanism of the photosensitizer was found to be in a way of apoptosis and necrosis on Hela cells since the apoptosis rate of ZnPc-(Lys)5at different time points were measured,early apoptosis rate at 0.5h,1h,2hand 4hwere(50.36±1.02)%,(40.80±0.72)%,(30.87±2.80)%,and(26.87±0.81)%,respectively.The late apoptosis rate were(34.23±0.55)%,(33.97±1.40)%,(45.57±1.12)%,and(45.57±1.22)%,respectively.The ultrastructure of Hela cells mainly showed early apoptosis after photodynamic treat for 1h,and showed late apoptosis and dying of bulging 2hlater.Conclusion ZnPc-(Lys)5induces death of Hela cells mainly through the apoptosis pathway.
