几种微囊藻定量方法的比较
Comparison of Several Methods for Microcystis Quantitation
【摘要】 以铜绿微囊藻单细胞藻株PCC7806和群体藻株XW01为实验材料,通过梯度稀释、细胞计数测定了藻细胞密度与OD值、叶绿素a含量、藻蛋白含量的线性关系曲线。结果显示720 nm处的OD值比其他短波长测定的OD值有更好的相关性。用4种不同氮浓度培养的微囊藻为材料,比较了OD、叶绿素a和蛋白质浓度3种定量方法的可靠性。结果发现OD法能更稳定地反映了微囊藻的浓度,而其他2个指标则由于细胞的不同生理状态差异较大。结果提示,OD720是用于测定微囊藻细胞浓度的最佳方法。
【Abstract】 Two Microcystis aeruginosa strains, unicellular strain PCC7806 and colonial strain XW01 were used to evaluated quantitative methods for cellular concentration. The linear relations of optic density value( OD), chlorophyll a and protein concentrations to cell densities were measured based on the gradient dilution and the cell counting. The results showed that the OD at 720 nm had a better correlation than that of the other short wavelength. To compare the reliability of the OD, chlorophyll a and protein concentration, different Microcystis cells cultured in four nitrogen concentrations were measured. Data displayed that OD at 720 nm was stably consistent with the cell concentration, while the other two indexes varied widely because of the different physiological status of the cells. The results indicated that OD720 was the best method used for the quantitation of Microcysits cell concentration.
