提取石油烃降解菌T1蛋白的有效方法研究
Petroleum Hydrocarbon Degrading Bacteria of T1 Protein Effective Extraction Method
【摘要】 该实验采用玻璃珠震荡、热裂解、反复冻融和超声波4种方法对细菌蛋白的提取、提取介质的裂解液。通过观察细菌裂解,蛋白浓度和蛋白SDS-PAGE电泳条带,得到了有效且稳定的石油烃降解菌蛋白质的提取方法。结果超声波效果最佳,热裂解和玻璃珠震荡效果一般,反复冻融和超声波提取蛋白的SDS-PAGE条带分布基本相同,其超声波获取蛋白丰度最高。蛋白质质量浓度测定显示,超声法提取蛋白质质量浓度最高。结果表明:超声法可高效提取石油烃降解细菌蛋白质,4种提取方法的稳定性和重复性均较好。
【Abstract】 In this experiment, the extraction medium lysates were used glass beads, thermal shock cracking and repeated freezing, thawing and ultrasound 4 methods bacterial protein extract, degree of fragmentation by observing bacteria, protein content and protein extracts by SDS-PAGE bands changes were compared the effect of different extraction method. So as to obtaine a stable, effective petroleum hydrocarbon degrading bacteria protein extraction methods, results, ultrasonic method works best thermal shock glass bead lysis method and the general effect, ultrasound and repeated freezing and thawing ex-tracting protein bands by SDS-PAGE distribution is basically the same, but ultrasonic Extraction protein bands highest abun-dance. Determination of the mass concentration of protein display, ultrasonic extraction highest concentration of protein qual-ity. Ultrasound method can effectively extract the petroleum hydrocarbon degrading bacteria protein, Four kinds of methods have good stability and reproducibility.
