miR-320a、miR-18b、miR-22和Let-7b在慢型克山病患者血清中表达及意义的研究

【摘要】 目的通过检测慢型克山病和对照组外周血中miR-320a、miR-18b、miR-22、Let-7b的表达水平,探讨其作为疾病鉴别诊断的生物标志物的意义。方法收集克山病病区慢型克山病患者(40例)、非病区对照组(40例)及病区对照组(40例)的血样标本,采用实时荧光定量PCR技术检测各组研究对象血清中目标miRNA的表达水平。结果 miR-320a、miR-18b和Let-7b在两对照组间比较有统计学意义(P<0.05);比较克山病组和病区对照组,miR-320a和Let-7b有统计学意义(P<0.05),提示这3个目标miRNA与高血压具有相关性。miR-22在三组比较中均无意义。结论 miR-320a、miR-18b、miR-22和Let-7b在克山病患者中未见显著表达,为慢型克山病的鉴别诊断提供了新思路。

【Abstract】 Objective To investigate the expression and significance of serum miR-320 a, miR-18 b, miR-22 and Let-7b in the patients with chronic Keshan disease( CKD) and the control groups.Methods 40 CKD and 40 cases of control were from the endemic areas of Keshan Disease, and another group of control( 40) from the non-endemic area. PCR method was used to detect the expression of serum miRNAs.Results The expression quantity of miR-320 a, miR-18 b and Let-7b between the two groups of control were showed statistically significant( P <0.05). miR-320 a and Let-7b were significant changes between the CKD and the control group from the endemic areas of Keshan Disease.The results suggest that miR-320 a, miR-18 b and Let-7b are related to hypertension. miR-22 among 3 groups had no statistical significance.Conclusions The target miRNAs were no significant in CKD, which provides ideas for the differential diagnosis of the disease.
