1,6-二磷酸果糖对甲状腺癌细胞凋亡和活性氧水平的影响
The Effect of fructose 1,6-bisphosphate on the Apoptosis and the Change of Reactive Oxygen Species in the Papillary Thyroid Carcinoma Cell
【摘要】 目的:研究不同浓度1,6-二磷酸果糖(F1,6BP)对人甲状腺乳头状癌细胞增殖、凋亡及活性氧(ROS)水平的影响。方法:F1,6BP体外孵育W3人乳头状甲状腺癌细胞株,MTS比色法检测细胞增殖、流式细胞术检测细胞凋亡、Western Blot法检测活化型caspase3表达,2-7-二氯氢化荧光素乙二脂(DCFH-DA)染色法结合荧光酶标仪检测细胞ROS水平,比色法分析细胞内氧化型谷胱甘肽(GSSH)含量。结果:2.5~10mmol/L F1,6BP可抑制W3细胞的增殖,诱导凋亡,呈浓度和时间依赖性变化。2.5~10mmol/L F1,6BP干预W3细胞48h后,细胞内ROS和GSSH含量逐渐升高,呈浓度依赖性变化。结论:F1,6BP能抑制人甲状腺乳头状癌W3细胞增殖,促使细胞凋亡,可能与细胞内ROS水平增加密切相关。
【Abstract】 Objetive:This study aims to investigate the apoptosis-inducing effect and the reactive oxygen species(ROS)level of fructose1,6-bisphosphate(F1,6BP)on the papillary thyroid carcinoma W3.Methods:W3cells were treated with F1,6BP in vitro.The changes of cell viability and cell nucleus morphology were examined by cell proliferation assay and Hoechst staining.Apoptosis levels of these cells were measured by flow cytometry,and the expression of caspase3 cleavage in cells was examined by western blot.The changes of ROS level and the percentage of oxidized glutathione in total glutathione in W3 cells were detected by 2′,7′-dichlorodihydrofluorescein diacetate(DCFH-DA)staining or colorimetry assay.Results:F1,6BP inhibited the growth of W3 cells and induced typical apoptosis of W3 cells significantly in dose and time manner.Intracellular ROS level and the percentage of oxidized glutathione in total glutathione were also increased in dose-manner after F1,6BP treatment.Conclusion:F1,6BP can inhibit the proliferation of W3 cells and induce the apoptosis of W3 cells through upregulating the generation of ROS.
