黄金茶提取物对中波紫外线损伤HaCaT细胞的保护作用研究
Photo-Protection of Huangjin Tea Extracts on HaCaT Cell From Ultraviolet B-Induced Damage
【摘要】 为了考察黄金茶提取物防治中波紫外线(UVB)对人表皮角质形成细胞损伤作用,采用不同浓度的黄金茶提取物对HaCaT细胞进行预处理6 h,采用60 m J·cm2的剂量照射细胞,以MTT法检测细胞生存率,吸取细胞上清液检测谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和乳酸脱氢酶(LDH)活性以及丙二醛(MDA)含量变化,用流式细胞仪检测细胞凋亡变化。结果表明,UVB辐射对HaCaT细胞造成严重损伤,黄金茶提取物可提高UVB照射后HaCaT细胞的存活率,提高GSH-Px、SOD活性,降低LDH活性和MDA含量,同时黄金茶提取物能够降低UVB所引起的细胞凋亡率升高(P<0.01),还可以减少UVB诱导HaCaT细胞损伤或凋亡,具有一定的光保护功能,其作用机制可能与清除氧自由基和增强抗氧化能力有关。
【Abstract】 In order to investigate the photo-protective effect of Huangjin tea extracts on HaCaT cells damaged from ultraviolet B radiation, sub-confluent HaCaT cells were incubated for six hours with different doses of Huangjin tea extracts, and then irradiated with 60 m J·cm2doses of UVB irradiation. The cell viability was detected by MTT method. The activity of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and lactate dehydrogenase(LDH), and the level of malondialdehyde(MDA) of supernatant were detected with colorimetric methods. Annexin V/PI double staining was performed to evaluate the change of apoptosis using flow cytometry.Results show the cell viability was inverse correlating with the irradiated dosage. Huangjin tea extracts could enhance the cell viability,SOD and GSH-Px activity of supernatant under UVB irradiation, decrease the activity of LDH and the content of MDA in dose-dependent manner. Compared with UVB irradiated model group, the apoptosis rate of pretreated HaCaT cell was decreased(P <0.01). Conclusion Huangjin tea extracts could relieve the oxidation cell damaged and apoptosis from UVB irradiation to HaCaT cells. Huangjin tea extracts had the function of photo-protection by clearing the oxyradical and raising the oxidase activity.
