金银花褐斑病病原检测和ITS鉴定
Pathogen detection and ITS identification of Lonicera japonica brown spot
【摘要】 为了提高金银花褐斑病防治的靶向性,加强对银花褐斑病病原的检测。依据科赫氏法则,采用‘巨花一号’金银花褐斑病病叶为试材,经过平板分离、菌种纯化、叶片接种试验和ITS鉴定。通过3次病原菌分离、检测,共分离真菌71株,从金银花叶片上首次筛查出番茄匍柄霉(Stemphylium solaini)、旋丝毛壳(Chaetomium bostrychodes)两种潜在致病菌;另外,还有2株具有生防价值的球毛壳菌(Chaetomium globosum)和黑附球菌(Epicoccum nigrum);试验证实拟茎点霉(Phomopsis sp.)为金银花褐斑病病原微生物。
【Abstract】 Leaf spot is one of the serious leaf diseases of honeysuckle,which has caused a lot of trouble to the honeysuckle production,so the pathogen detection of honeysuckle leaf spot has guiding significance for improving the target control.According to Koch’s rules,leaves of Lonicera japonica(‘Juhua No.1’)which had been infected by leaf spot were picked and cultured in laboratory.Separation and purification of the microbial species were conducted using plate screening technique.The leaf inoculation experiment and ITS identification were studied later.Through two years of culture and three times of detachment and detection,71 fungus strains were separated,among which two kinds of potential pathogens,Stemphylium solaini and Chaetomium bostrychodes,were first screened out.In addition,two other strains with biocontrol value named as Chaetomium globosumand Epicoccum nigrum were found.The experiment proved that Phomopsis sp.was the pathogenic microorganism of honeysuckle leaf spot.
