针灸对环磷酰胺化疗小鼠骨髓细胞DNA含量动态变化的影响

【摘要】 目的:通过观察针灸对环磷酰胺(CTX)化疗小鼠骨髓细胞DNA含量的影响,探讨针灸改善化疗后骨髓抑制、升高外周血白细胞的分子生物学机制。方法:将昆明种雄性小鼠160只按体质量随机分为正常组、模型组、针刺组、艾灸组,每组40只。用CTX建立骨髓抑制模型。模型成功后,每天定时对针刺组、艾灸组小鼠"大椎""膈俞""肾俞""足三里"给予针刺、艾灸治疗,连续6d。正常组、模型组每日陪同抓取固定,不治疗。镜检外周血白细胞数、流式细胞仪检测各组小鼠骨髓细胞G1期、G2期DNA含量,进行数据统计分析。结果:针刺和艾灸可使CTX小鼠外周血白细胞提前1天回升并提前1天超过基础水平;针刺和艾灸可以上调骨髓细胞G1期、G2期DNA含量,在治疗第5天针刺组、艾灸组骨髓细胞G1期、G2期DNA含量高于模型组,差异具有统计学意义(P<0.05)。结论:针刺和艾灸可以通过上调CTX小鼠骨髓细胞G1期、G2期DNA的含量,加速骨髓细胞DNA的修复,改善化疗所致骨髓细胞增殖、分裂受抑制的状态,发挥提升外周血白细胞,维护造血功能的作用。

【Abstract】 Objective:To explore the mechanism underlying acupuncture and moxibustion improving suppression of bone marrow and periphery leucocyte by observation of effects of acupuncture and moxibustion on DNA content during different mitotic cycles of bone marrow cells in cyclophosphamide-induced mice.Methods:Total of 160 Kunming mouse were randomly divided into control group,model group,acupuncture group and moxibustion group,40 mice in each group.The model of myelosuppression was induced by cyclophosphamide.Mice in the acupuncture group and moxibustion group were respectively given the treatment of acupuncture and moxibustion on acupoints including ‘Dazhui’(GV 14),‘Geshu’(BL 17),‘Shenshu’(BL 23) and ‘Zusanli’(ST 36),for 6 consecutive days.The mouse in the control group and model group were subjected only i xation but no extra treatment.The number of periphery leucocyte was recorded by microscopy.The DNA content in G1 and G2 phase of bone marrow cells was detected by l ow cytometry.Results:The recovery of periphery leucocyte in cyclophosphamide-induced mice was advanced by the treatment of acupuncture and moxibustion for one day,and the time of when number of periphery leucocyte exceeded the basic level was advanced for one day.In the 5th day of the treatment,the DNA content of G1 and G2 phase in the acupuncture and moxibustion groups were higher than those in the model group,indicating that acupuncture and moxibustion increased the DNA content of G1 and G2 phase(P<0.05).Conclusion:Acupuncture and moxibustion promote DNA repairmen by up-regulating DNA content of bone marrow cells in cyclophosphamide-induced mice,improving suppression of bone marrow and periphery leucocyte and recovering hematopoietic function.
