西藏沙棘茎尖组织快繁技术初步研究

【摘要】 本文以幼嫩茎尖为外植体,对西藏沙棘进行了组织培养成苗技术研究。结果表明,西藏沙棘茎尖生长最适宜的基本培养基为1/2 MS;茎尖生长最佳生长调节剂浓度配比为:6-BA 1.0 mg/L+NAA 0.l mg/L;蔗糖浓度为2.0mg/L时西藏沙棘茎尖生长苗高均值最大,达到2.82 cm。茎尖在增殖生长过程之中,适当的转接周期能改善芽苗枯死现象。转接周期为15 d时,芽苗枯死率最低为16.33%;当转接周期延长至30 d时,芽苗枯死率明显升高,达到52.67%。

【Abstract】 In this paper young and tender stem tips were used as explants to study tissue culture techniques for growing seedlings of Hippophae thibetana. The results showed that the optimal basic culture medium is 1/2 MS; the optimal growth regulator concentration ratio for Hippophae thibetana stem tips growth is 6-BA1.0mg.L-l +NAA0.lmg.L-l; The mean stem-tip height increment reached maximum of 2.82 cm with sucrose concentration of 2.0mg.L-1; In the stem-tip proliferation process, appropriate switching cycles could lessen dieback of bud seedlings; The dieback rate reached minimum of 16.33% with the replanting interval of 15 d and increased significantly to 52.67% when the replanting interval extended to 30 d.
