胃炎饮方对胃溃疡大鼠表皮生长因子和转化生长因子β1表达的影响
Influence of Weiyanyin Formula on the Expression of Epidermal Growth Factor and Transforming Growth Factor-β1 in Gastric Ulcer Rats
【摘要】 目的从调节表皮生长因子(EGF)和转化生长因子β1(TGF-β1)表达的角度探讨胃炎饮方促进胃溃疡愈合的分子机制。方法 50只Wistar大鼠随机抽取8只作为假手术组,其余42只采用冰醋酸腐蚀法建立胃溃疡大鼠模型。将造模成功的32只分为模型组、胃高组、胃低组、对照组,每组8只。胃高组、胃低组分别灌服生药浓度为2.14、1.07 g/ml胃炎饮煎剂10 ml/kg,对照组灌服浓度为0.368 mg/ml的奥美拉唑液10 ml/kg。各给药组从造模术后第3天开始灌胃,假手术组、模型组灌胃等量生理盐水,每日1次,共14天。双抗体夹心ABC-ELISA法检测各组大鼠EGF、TGF-β1水平,免疫组化方法检测各组大鼠EGF、TGF-β1表达,RT-PCR方法检测各组大鼠EGF mRNA、TGF-β1mRNA的表达。结果与假手术组比较,模型组大鼠胃组织EGF、TGF-β1水平显著升高,平均光度值显著升高,而平均灰度值显著下降,EGF mRNA、TGF-β1mRNA表达显著升高(P<0.05或P<0.01);与模型组比较,各治疗组EGF、TGF-β1水平及平均光度值显著升高,平均灰度值显著下降,EGF mRNA、TGF-β1mRNA表达显著升高,溃疡面积均明显缩小(P<0.05或P<0.01);与对照组和胃低组比较,胃高组EGF、TGF-β1水平及平均光度值显著升高(P<0.05或P<0.01),EGF mRNA、TGF-β1mRNA表达及溃疡面积差异具有统计学意义(P<0.05或P<0.01)。结论胃炎饮可促进胃溃疡大鼠胃组织EGF和TGF-β1表达,加强胃黏膜修复,可能是提高溃疡愈合的可能机制。
【Abstract】 Objective To explore the molecular mechanism of Weiyanyin Formula( WYYF) in promoting the healing of gastric ulcer through regulating the expression of epidermal growth factor( EGF) and transforming growth factor-β1( TGF-β1). Methods Chosen randomly from 50 Wistar rats,8 rats were used as sham-operation group and the remaining rats were made into gastric ulcer models by freezing acetic acid corrosion. The 32 successful models were then divided into model group,high dose group,low dose group and control group( n = 8). The high dose group and low dose group were administered with 10 ml / kg WYYF at the concentration of 2. 14 and 1. 07 g / ml by gavage,while the control group were administered 10 ml / kg omeprazole solution at the concentration of 0. 368 mg / ml.Each treated group began gastric administration 3 days after modeling. The sham-operation group,model group were given normal saline once a day for 14 days. The levels of EGF,TGF-β1 were detected by double-antibody sandwich ABC-ELISA assay and the expression of EGF,TGF-β1 were tested by immunohistochemistry. The RT-PCR method was applied to detect the mRNA expression of EGF and TGF-β1 of each group of rats. Results Compared with the sham-operation group,the content of EGF and TGF-β1 and the average luminosity value of the model group was significantly higher,with the average gray scale lower and the expression of mRNA of EGF and TGF-β1 significantly higher( P < 0. 01 or P < 0. 05). Compared with the model group,the content and the average luminosity value of EGF and TGF-β1 in every treatment group was significant higher,with the average gray scale lower,the expression of mRNA higher and ulcer area smaller( P < 0. 01 or P < 0. 05). Compared with the control group and the low dose group,the levels of EGF and TGF-β1 and average luminosity value in the high dose group were significantly higher( P < 0. 05 or P < 0. 01),and the difference in the expression of EGF mRNA and TGF-β1 mRNA and ulcer area was statistically significant( P < 0. 05 or P < 0. 01). Conclusion WYYF could promote the expression of EGF and TGF-β1 in gastric tissue of gastric ulcer rats and strengthen the recovery of gastric mucosa,which may be the possible mechanism of its role in ulcer healing.
