基于细胞凋亡研究桔梗元参汤含药血清对炎性气道黏液高分泌的抑制作用

【摘要】 目的:探讨桔梗元参汤含药血清对气道黏液高分泌的作用和相关分子机制。方法:培养A549细胞株,构建气道黏液高分泌模型,以人中性粒细胞弹性蛋白酶为刺激因素,采用血清药理学方法制备含药血清,观察细胞内活性氧含量以及Bcl-2、Bax、Caspase3和MUC5AC的蛋白表达。结果:与模型组相比,桔梗元参汤含药血清(5~20%)预处理后ROS含量由78.1降至39.2;10%和20%梗元参汤含药血清能够显著上调Bcl-2的蛋白表达,同时下调Bax、caspase 3和MUC5AC蛋白表达。结论:桔梗元参汤含药血清具有抑制气道黏液高分泌的作用,其作用机制可能与调控MUC5AC和caspase家族蛋白表达有关。

【Abstract】 Object: To investigate the effect and mechanism of Jiegengyuanshen decotion containing serum in the airway inflammation. Method:The cell model of mucous hypersecretion was made by human lung A549 cell stimulated by human neutrophil elastae( HNE). Serum pharmacology method was used to detect reactive oxygen species level and Bcl-2,Bax,Caspase3 and MUC5 AC protein expression. Results: Comparing with the model group,Jiegengyuanshen decotion containing serum decrease the level of ROS from 78. 1 to 39. 2( P < 0. 05); Different dose of Jiegengyuanshen decotion containing serum( 5 ~ 20%) significantly upregulated Bcl-2 expression,while,downregulated Bax,Caspase 3 and MUC5 AC expression. Conclusion: Jiegengyuanshen decotion could inhibit the airway inflammation,and its mechanism maybe related to regulate MUC5 AC and caspase proteins expression.
