银丹心脑通软胶囊通过促进NO释放抑制球囊损伤所致大鼠血管内膜增生

【摘要】 目的研究银丹心脑通软胶囊(YDXNT)对球囊损伤所致大鼠血管内膜增生的影响并探讨其机制。方法将雄性SD大鼠随机分为假手术组(Sham)、模型组(Model)、YDXNT低剂量组(YDXNT-L)和YDXNT高剂量组(YDXNT-H),行颈总动脉球囊损伤造模后次日开始灌胃给药,YDXNT低、高剂量组大鼠分别给予YDXNT 0.5和1.0 g/kg,每日一次;Sham组与Model组大鼠给予生理盐水,共14 d。HE染色光镜观察血管形态学变化,RT-PCR法检测其e NOS mRNA的表达,比色法检测大鼠血浆中NO含量,免疫酶联吸附试验(Elisa)检测血浆环磷酸鸟苷(c GMP)的含量。结果 YDXNT能显著减轻球囊损伤所致颈动脉管腔狭窄,并可上调血管e NOS mRNA的表达及升高血浆中NO、c GMP(P<0.01)含量。结论 YDXNT可抑制因球囊损伤所致大鼠的颈动脉血管内膜增生,其机制可能与YDXNT促进NO释放有关。

【Abstract】 Objective To investigate the effects of Yindan Xinnaotong capsule( YDXNT) on intimal hyperplasia in a rat model of carotid artery balloon injury,and explore its possible mechanisms. Methods Male Sprague Dawley rats were randomly divided into 4 groups,Sham,Model,YDXNT- L( 0. 5 g / kg) and YDXNT- H( 1. 0 g /kg). The rat carotid arterial intima- injured model was established with balloon- injured method,YDXNT were administered once daily for 14 days in the YDXNT groups,and volume- matched normal saline were administered in the Sham and the Model groups. After 14 days,the neointimal hyperplasia level was evaluated by hematoxylin- eosin staining. The expression of e NOS mRNA in carotid artery of rats were examined by RT- PCR,plasma NO and c GMP content were detected by colorimetric and immune enzyme- linked assay respectively. Results Compared with those in the model group,the carotid artery lumen stenosis induced by balloon injury were remarkably alleviated in YDXNT 0. 5 g / kg and 1. 0 g / kg groups. Moreover,the expression of e NOS mRNA was up- regulated,while the plasama NO,c GMP contents increased remarkably. Conclusion YDXNT could suppress the vascular neointimal hyperplasia induced by balloon injury,which related to up- regulation of NO production.
