江苏地区7株H9N2亚型禽流感病毒HA基因的序列分析

【摘要】 目的了解江苏地区H9N2亚型禽流感病毒的遗传变异情况及分子生物学流行规律。方法采用逆转录聚合酶链反应(RT-PCR)技术对7株从江苏淮安地区分离的H9N2亚型禽流感病毒的血凝素(HA)基因进行扩增和测序,并对所获得的HA基因序列与该亚型的代表毒株进行同源性分析,构建系统进化树。结果 7个分离株核苷酸同源性达97%以上,与临近省份分离毒株同属国内常见的BJ94-like亚系。7株毒株HA裂解位点的氨基酸序列均为RSSR↓GLF,为低致病性禽流感病毒。结论 2014年江苏省H9N2禽流感病毒进化相对比较稳定,此7株毒株可能有着相同的起源。

【Abstract】 [Objective] To explore the status of genetic variation and law of molecular epidemiology of H9N2 subtype avian influenza viruses in Jiangsu Province.[Methods] Seven strains of avian influenza virus subtype H9N2 were isolated from Huai’an of Jiangsu Province,the whole hemagglutinin(HA) fragments of the viruses were amplified by reverse transcription polymerase chain reaction(RT-PCR),and the sequences were analyzed on homology and heredity evolution after the sequencing of the products. The phylogenetic tree was established. [Results] Homology analysis showed that the homology of HA nucleotide sequences was 97% above for different strains,and they belonged to the same major branch of the A/Chicken/Beijing/1/94(BJ94-like)in the phylogenetic tree. The amino acid motif of cleavage sites for all seven virus in the HA gene were RSSR↓GLF,which was consistent with the characterization of the low pathogenic avian influenza virus(LPAIV).[Conclusion] H9N2 subtype influenza viruses might have a stable evolution and common origin in Jiangsu in 2014.
