正肝汤对肝硬化大鼠肠道紧密连接蛋白OCLN和ZO-1表达的影响

【摘要】 目的:探讨正肝汤对肝硬化大鼠肠道紧密连接蛋白OCLN和ZO-1表达的影响。方法:30只SD大鼠随机分为正常组、模型组和正肝汤组,每组各30只。制备四氯化碳肝硬化大鼠模型,正肝汤组大鼠予正肝汤(1.5 g/100 g·d)灌胃干预。对肝组织进行苏木素-伊红染色,透射电镜观察小肠组织的超微结构,免疫印迹法检测小肠组织中OCLN和ZO-1的蛋白表达。结果:与正常组相比较,模型组大鼠的肝组织中胶原纤维广泛增生,纤维间隔相互连接,徦小叶形成,电镜见小肠黏膜上皮细胞表面微绒毛稀疏,减少、变短,排列不规则,部分断裂扭曲,脱落明显。正肝汤组大鼠的肠黏膜损伤较模型组明显减轻。正常组大鼠小肠组织中OCLN和ZO-1的表达量分别是模型组2.94和3.91倍,均P<0.01,差异显著。正肝汤组的小肠组织中OCLN和ZO-1的表达量分别是模型组1.74和1.91倍,与模型组相比均P<0.05,有统计学差异。结论:正肝汤能显著减轻肝硬化大鼠的肠黏膜损伤,并上调小肠组织中OCLN和ZO-1的蛋白表达。

【Abstract】 Objective: To investigate the effect of Zhenggan Decoction on OCLN and ZO-1 expressions in intestine of CCl4-induced cirrhotic rats. Methods: 30 Sprague-Dawley rats were randomly divided into 3 groups: control group( n= 10),model group( n = 10),Zhenggan Decoction group( n = 10). Cirrhosis was induced by subcutaneous injection of CCl4 administration of alcohol. The rats in Zhenggan Decoction group were intragastrically administrated with Zhenggan Decoction at a dose of 1. 5 g /( 100 g·day). The morphologic change of liver was evaluated by Hematoxylin-eosin staining. The ultra-structure of intestinal tissue was observed by transmission electron microscope. The expressions of OCLN and ZO-1 in intestine were detected by western blotting. Results: Compared with control group,collagen deposition,significant fibrous septa and cirrhotic nodules were observed in the liver of rats in model group. Electron microscope examination showed sparse and short microvilli of irregular arrangement on the surface of intestinal epithelial cells in cirrhotic rats. Some of the microvilli were distorted and dropped remarkably. The severity of intestinal mucosal damage in rats treated with Zhenggan Decoction was significantly reduced compared to that in cirrhotic rats. The expressions of OCLN and ZO-1 in intestinal tissue of control rats were significantly higher than that of cirrhotic rats with 2. 94-fold and 3. 91-fold over model group respectively( both P < 0. 01). The expressions of OCLN and ZO-1 in intestinal tissue of rats treated with Zhenggan Decoction were significantly increased and were 1. 74-fold and 1. 91-fold over that of cirrhotic rats( both P < 0. 05). Conclusion: Zhenggan Decoction significantly relieved the intestinal mucosa damage and up-regulated the expressions of OCLN and ZO-1 in intestinal tissue in cirrhotic rats.
