巴戟天与雌激素对骨质疏松大鼠破骨细胞RANK和CAII的表达影响分析
The analysis on Expression Effect of Morinda Root and Estrogen on Osteoporosis rat Osteoclast RANK and CAII
【摘要】 目的:探讨巴戟天与雌激素对骨质疏松大鼠破骨细胞RANK和CAII的表达影响。方法:选用4月龄SPF级健康成年雌性SD大鼠,建立去势大鼠骨质疏松模型,分离大鼠原代破骨细胞并分为3组:B组给予17β-雌二醇10-6mmol/L,C组给予1.0 mmol/L巴戟天,D组给予17β-雌二醇10-6mmol/L和1.0 mmol/L巴戟天进行培养,观察比较各组CAII、RANKmRNA表达等指标差异。结果:B、C和D组破骨细胞数量均较A组明显减少,而D组减少最明显(P<0.05);B、C、D组骨吸收陷窝面积相均低于A组(P<0.05),而D组骨吸收陷窝面积明显低于其他组(P<0.05);B、C、D组CAII、RANKmRNA基因表达水平明显低于A组(P<0.05),B组和C组RANKmRNA基因表达水平差异无统计学意义(P>0.05),而CAIImRNA基因表达水平差异有统计学意义(P<0.05),D组CAII、RANKmRNA基因表达水平最低(P<0.05)。结论:巴戟天联合雌激素能明显降低骨质疏松大鼠破骨细胞RANK和CAII的表达,起到抑制骨质疏松的作用。
【Abstract】 Objective: To discuss the expression effect of Morinda Root and estrogen on Osteoporosis rat Osteoclast RANK and CAII. Method: Choose 4 months SPF health adult female SD mice,set up castrate rat Osteoporosis model,separate rat original Osteoclasts and divided into three groups: group B was given 17 β- estradiol 10- 6mmol / L,group C was given Morinda Root 1. 0 mmol / L,group D was given 17 β- estradiol 10- 6mmol / L and Morinda Root 1. 0 mmol / L to cultivate,observe and compare the difference of CAII and RANKmRNA expression of groups. Result: Osteoclast amount of group B,C and D were less than A group obviously,while group D decrease was the most obvious( P < 0. 05); group B,C and D bone resorption lacuna area were less than that of group A( P < 0. 05),while group D bone resorption lacuna area was less than that of other groups( P < 0. 05); CAII and RANKmRNA gene expression level of group B,C and D were less than A group obviously( P < 0. 05),RANKmRNA gene expression level of group B and C had no significant meaning( P >0. 05),while CAIImRNA gene expression level had the significant meaning( P < 0. 05),CAII and RANKmRNA gene expression level of group D was the lowest P < 0. 05). Conclusions: Morinda Root and estrogen can reduce CAII and RANK gene expression of Osteoporosis rat Osteoclast obviously,having the effect of inhibiting osteoporosis.
