白芷香豆素类成分的提取及大孔树脂纯化的工艺研究
Optimization of Extraction and Purification Technology of Total Coumarins in Radix Angelicae Dahuricae with microporous resin
【摘要】 目的:研究白芷香豆素的最佳提取工艺及大孔树脂的最佳纯化工艺。方法:选用L9(34)进行正交设计试验,对白芷的醇提工艺进行优选,以欧前胡素的含量为指标,考察乙醇浓度、提取次数、提取倍数对提取工艺的影响。采用单因素试验法对影响白芷香豆素纯化效率的因素进行优化,采用HPLC法测定欧前胡素的含量。结果:得到的最佳醇提工艺条件为使用10倍量90%的乙醇,提取2次,每次2 h;最佳纯化工艺为:选定D101大孔树脂,上样浓度为每m L含生药0.3 g,最大上样量为7倍BV(7倍柱体积),上样速度为4BV·h-1,用6 BV的水以4 BV·h-1的速度洗脱,弃去洗脱液,再用7 BV的90%乙醇以4 BV·h-1的流速洗脱,收集洗脱液。结论:优选出醇提工艺稳定可靠,有效成分含量高;优选出的大孔树脂纯化工艺简便、稳定,有效成分转移率高。
【Abstract】 Objective: To optimize extraction and purificatin technology of total coumarins in radix Angelicae Dahuricae. Method: The extracting conditions were optimized by using L9( 34) orthogonal test. Employing the content of imperatorin as index,the effects of the concentration of ethanol,the time and cycles of extracting on the extracting process were investigated. Mono factor test was employed to investigate the main parameters which may affect the purification procedures with the concentration of imperatorin. The content of the imperatorin was verified by HPLC. Result: The most favorable conditions of extraction were 10 times amount of 90% ethanol for 2 cycles and every extracting time was 2 hours. The optimal purification procedure was using D101 macroporous resin. The content of coumarin was 0. 3 g m L- 1; the volume was 7 BV; the flow rate was 4 BV· h- 1. To elute with distilled water,the volume was 6 BV and the flow rate was 4 BV· h- 1,giving up the eluate. To elute with 90% ethanol,the volume was 7 BV and the flow was 4 BV· h- 1,gathering the elute. Conclusion: Optimum conditions of ethanol extraction were obtained and the optimized extraction technology was stable and efficient. The amount of active component was high. The optimized purification technology was suitable of coumarin for its simple and high effectiveness.
