利用基因表达芯片筛选与无功能垂体腺瘤侵袭性相关基因的研究

【摘要】 目的探寻与无功能垂体腺瘤(NFPA)侵袭性相关基因。方法选择7例NFPA和3例正常垂体组织进行基因表达芯片分析。通过生物信息学和统计生物软件,筛选出与NFPA侵袭性相关的基因。进一步利用43例NFPA标本,通过QPCR和免疫组织化学方法验证筛选出的ENC1基因与侵袭性NFPA的相关性。结果在侵袭性NFPA、非侵袭NFPA和正常垂体组织三组之间,筛选出共表达发生变化的基因56个,其中在侵袭性NFPA中35个基因上调和21个基因下调。QPCR结果显示:ENC1在侵袭性NFPA表达低于非侵袭性NFPA表达(P=0.010)。免疫组化结果显示:ENC1在正常垂体组织呈强阳性,而在侵袭性NFPA呈弱阳性。结论 ENC1可能是侵袭性NFPA的一个重要生物标记。

【Abstract】 Objective To explore the invasion-associated gene of non-functioning pituitary adenomas( NFPA). Methods Gene expression microarray analysis was performed in 7 NFPA patients and 3 subjects with normal pituitary tissue. The invasion-associated genes of NFPA were selected through bioinformatics and statistical biological software. The correlation of ENC1 gene with aggressive NFPA was verified by QPCR and immunohistochemistry in 43 NFPA samples. Results A total of 56 genes were found to be related to the invasiveness in invasive NFPA, non-invasive NFPA and normal pituitary glands. Of them, expressions of 35 genes were up-regulated and those of 21 down-regulated in invasive NFPA compared with non-invasive NFPA and normal pituitary glands. The ENC1 expression level was significantly lower in invasive NFPA compared with non-invasive NFPA(P = 0.010). Immunohistochemistry showed ENC1 was strongly positive in normal pituitary tissues and weakly positive in invasive NFPA. Conclusions ENC1 may be an important biomarker for invasive NFPA.
