脉冲射频对保留性神经损伤大鼠Nav1.8 mRNA表达的影响

【摘要】 目的:通过观察保留性神经损伤(spared nerve injury,SNI)模型大鼠接受脉冲射频(pulsed Tadiofrequency,PRF)治疗后疼痛行为学和背根神经节(dorsal root ganglion,DRG)电压门控钠离子通道1.8(Nav1.8)m RNA的表达水平,探讨PRF治疗神经病理性疼痛的可能机制。方法:选择SD大鼠32只,随机均分成4组:空白对照组(C组)、模型组(SNI组)、假治疗组(Sham组)和治疗组(PRF组)。于制模前0天,制模后4、6、8、10天,PRF治疗后6、12、18、24、30、36、42天测定四组大鼠机械缩足阈值(mechanical withdrawal threshold,MWT);在PRF治疗6天后每组各处死2只大鼠,42天后每组处死剩余大鼠,取其腰4、5的背根神经节,采用逆转录聚合酶链反应(RT-PCR)半定量分析各组大鼠背根神经节Nav1.8 m RNA的表达情况。结果:SNI术后,SNI组、Sham组和PRF组大鼠的MWT较C组显著降低(P<0.01);PRF治疗后,PRF组大鼠的MWT及大鼠背根神经节Nav1.8 m RNA相对表达量显著高于SNI组和Sham组(P<0.01)。结论:PRF治疗对SNI神经病理性疼痛大鼠具有镇痛作用,Nav1.8可能是PRF发挥镇痛作用的一个关键靶点。

【Abstract】 Objective: To investigate the behavioral changes and the dynamic expression of Nav1.8 m RNA in the dorsal root ganglion(DRG) of rats in spared nerve injury after pulsed radiofrequency treatment. Methods: Thirty-two SD rats were randomly divided into four groups: control group(C group), model group(SNI group), sham treatment group(Sham group) and PRF treatment group(PRF group). The mechanical withdrawal threshold(MWT) was measured before SNI surgery, after SNI surgery in 4,6,8,10 days, and after PRF treatment in 6,12,18,24,30,36,42 days, respectively. The expression of Nav1.8 m RNA in dorsal root ganglion was quantified by reverse transcription polymerase chain reaction in each group at 6, 42 days after PRF treatment, respectively. Results: The MWT of SNI group, Sham group and PRF group were significantly lower than that in control group after SNI surgery(P < 0.01). The MWT and the expression of Nav1.8 m RNA of PRF group were significantly higher than those of SNI group and Sham group after PRF treatment(P < 0.01). Conclusion: PRF provides an effective treatment for SNI neuropathic pain rats, which might be regulated through the expression of Nav1.8.
