2012-2014年我国南方地区猪流行性腹泻病毒遗传进化分析

【摘要】 为分析猪流行性腹泻病毒(PEDV)的遗传变异情况,本试验对2012-2014年我国南方地区7个省市17份PEDV阳性样品的S基因主要的中和表位区(COE)序列进行了RT-PCR扩增、克隆和测序,并与国内外的代表性毒株进行序列比对和遗传进化分析,结果表明,本研究中的17株:PEDV毒株COE中有16株属于第1大群,而GD/FSss/2014属于第Ⅱ大群,所有COE的核苷酸和氨基酸同源性分别为92.9%~99.8%和88.6%~100%,它们与经典毒株CV777的核苷酸和氨基酸同源性分别为93.3%~99.5%和90%~98.6%,与疫苗株CV777 Vaccine的核苷酸和氨基酸同源性分别为95%~97.1%和92.9%~97.9%。COE的氨基酸序列发生了不同程度的点突变,为PEDV遗传进化分析和疫病防制提供了新的参考。

【Abstract】 In this study,in order to investigate the variation in the core neutralizing epitope(COE) of S gene of porcine epidemic diarrhea virus(PEDV),a total of 17 PEDV positive samples were collected from different breeding farms from 7 provinces in the southern region of China during 2012 to 2014,and the COEs were amplified by RT-PCR,cloned and sequenced.Sequence analysis and genetic variation of 17 COEs were compared with other PEDV reference strains selected from GenBank.Our results showed that 16 of 17 strains were closely related to each other and belonged to the first group,and the GD/FSss/2014 strain belonged to the second group.Sequence analysis showed that the COE of 17 strains shared 92.9%—99.8% nucleotide and 88.6%—100% deduced amino acid identities to each other,and exhibited 93.3%~99.5% nucleotide and 90%—98.6% deduced amino acid identity with the reference PEDV strain of CV777.Compared with the CV777 vaccine strain,the strains sequenced in this study had 95-97.1% nucleotide and 92.9~97.9% deduced amino acid identity.Meanwhile,there were some amino acid mutations in the COE.This study could provide the theoretical foundation for selection and further study of comprehensive prevention and control of PEDV.
