一种山东地栽香菇病害病原菌的分离及鉴定
Isolation and Molecular Identification of a Pathogen in Lentinula edodes
【摘要】 从山东肥城采集到香菇病害样品,分离得到病原菌分离物FC,提取其基因组总DNA,利用真菌通用引物ITS1/ITS4扩增其r DNA ITS区域序列并进行测序。将获得的序列进行同源性比对和构建系统进化树,发现FC分离物与Gen Bank中8个米根霉(R.oryzae)分离物同源性最高,为98.27%,结合形态学特征,证明该分离物为米根霉(R.oryzae)。
【Abstract】 A fungi strain, FC, was isolated from Lentinula edodes and its genome total DNA was obtained. The internal transcribed spacers(ITS) was amplified by PCR using fungi universal primers ITS1 and ITS4. Homology analysis and constructed phylogenetic tree showed that FC shared nucleotide identity of 98.27% with eight Rhizopus oryzae isolates. Combined morphological characteristics, the FC was R. oryzae.
