分枝虫草液体培养基筛选及其菌丝成分分析

【摘要】 以分枝虫草(Cordyceps ramosa)菌丝生物量作为指标,研究不同碳源和氮源对分枝虫草液体培养的影响,采用正交设计试验法筛选最佳液体培养基配方,并分析菌丝主要营养成分。结果表明,分枝虫草液体培养基的最佳碳源为甘油,最佳氮源为硝酸钾,最佳配方是:甘油35 g·L-1、硝酸钾12 g·L-1、酵母浸膏12 g·L-1、麦芽浸出物10 g·L-1、H2O 1 L;其菌丝的粗蛋白、粗脂肪和总灰分含量分别为32.90%、1.20%和5.89%,其腺苷、多糖和虫草酸的含量分别为0.805%、5.89%和5.61%;含有17种氨基酸,总含量为23.97%;微量元素铁、锰、铜、锌和硒含量分别为76.70 mg·kg-1、5.10 mg·kg-1、3.70 mg·kg-1、33 mg·kg-1和0.176 mg·kg-1,重金属铅、砷、汞的含量均符合国家食用菌相关标准。分枝虫草的液体培养可作为分枝虫草开发利用的重要途径。

【Abstract】 Effects of carbon and nitrogen sources on the optimization of liquid-culture medium for Cordyceps ramosa mycelial biomass, and the optimum liquid medium screening by the orthogonal design, and the main components of mycelium were analyzed. The results showed that the optimal carbon and nitrogen sources were glycerol and KNO3 respectively, and the optimal medium were glycerol 35 g·L-1, KNO312 g·L-1, yeast extract fermentation 12 g·L-1, malt extract 10 g·L-1and H2 O 1 L. The mycelium contained crude protein(32.9%), crude fat(1.2%) and ash(5.89%). The contents of adenosine, polysaccharide,and cordycepic acid were 0.805%, 5.89% and 5.61%. And 17 kinds of amino acids(23.97%) were detected from the mycelium. The microelement levels, such as Fe, Mn, Cu, Zn, Se, were 76.70 mg·kg-1, 5.10 mg·kg-1, 3.7 mg·kg-1, 33 mg·kg-1and0.176 mg·kg-1, respectively. Levels of the heavy metals, such as Pb, As and Hg, met the edible fungus safety standard levels specified by the Ministry of Health of the People’s Republic of China. The results could provide important way for the development and utilization of C. ramosa.
