高效液相色谱法测定窝儿七中鬼臼毒素和4’-去甲基鬼臼毒素的含量

【摘要】 目的建立高效液相色谱法测定窝儿七中鬼臼毒素和4’-去甲基鬼臼毒素的含量。方法色谱柱为COSMOSIL-C18(4.6 mm×250 mm,5μm),柱温25℃;以甲醇为流动相A,0.4%磷酸溶液为流动相B进行梯度洗脱;流速1 m L·min-1;检测波长330 nm。结果 4’-去甲基鬼臼毒素、鬼臼毒素的线性范围分别为4.5~224.0μg·m L-1(r=0.999 2)和20.0~1 000.0μg·m L-1(r=0.999 4),平均回收率分别为99.8%(RSD=1.5%)和99.8%(RSD=1.7%)。结论本方法灵敏、准确、重复性好,为窝儿七的质量控制提供参考依据。

【Abstract】 Objective To determine the content of podophyllotoxin and 4’-demethylpodophyllotoxin in Diphylleia sinensis. Li by HPLC. Methods The COSMOSIL-C18 column(4.6 mm×250 mm, 5 μm) was used. The column temperature was 25℃. Methanol-0.4% glacial acetic acid solution was used as the mobile phase at 1 m L·min- 1. The detection wavelength was 330 nm. Results The linearity of 4’-demethylpodophyllotoxin and podophyllotoxin was 4.5-224.0 μg·m L-1(r = 0.999 2) and 20.0- 1 000.0 μg·m L-1( r = 0.999 4), and the average recoveries were 99.8%(RSD= 1.5%) and 99.8%(RSD = 1.7%). Conclusion The method is sensitive, accurate and reproducible. It can be used for the determination of main compositions in Diphylleia sinensis. Li.
