白纹草的组织培养和快速繁殖技术研究

【摘要】 为提高白纹草(Chorophytum bichetii)种苗质量和繁殖效率,以其顶芽或茎段为外植体材料,比较不同实验时间外植体的灭菌效果,筛选诱导丛生芽和继代繁殖的最佳激素配比,建立白纹草离体快繁的技术体系。结果表明,3月取材,外植体的灭菌效果最好,顶芽和嫩茎段的灭菌率分别为74.7%和82.7%;以MS为基本培养基,附加6-BA 2 mg/L+IBA 0.2 mg/L为最佳诱导丛生芽的培养基;以6-BA 2 mg/L+KT 0.2 mg/L为最佳继代增殖培养基,增殖系数达5.1;使用较高浓度6-BA时会发生较严重的玻璃化现象。以1/2MS+NAA 0.1 mg/L为最佳生根培养基,生根率100%;根长0.5 cm左右时最适宜移栽。用泥炭和珍珠岩按2:1混合的基质移栽,移栽成活率达99%以上,并且种苗健壮。在保证繁殖系数前提下尽量降低细胞分裂素浓度,并在继代中不断进行筛选剔除变异株。该技术体系能够满足规模化生产的要求。

【Abstract】 The techniques of the tissue culture and rapid propagation of Chorophytum bichetii were studied.The quality and the induction rates were higher by optimizing test periods and the mediums. The resultsshowed that the stems and terminal buds as explants could be induced to sterilized shoots. The sterilization ratein March was higher, and the stems and terminal buds were 74.7% and 82.7%. With MS as basic culturemedium, different concentrations of plant growth regulator 6- BA and NAA, KT were added to induction,proliferation and rooting. The results indicated that 6-BA 2 mg/L+ IBA 0.2 mg/L was the suitable medium forexplants induction of cluster buds; 6-BA 2 mg/L+ KT 0.2 mg/L was the best medium for multiplication, andthe multiplication coefficient could reach up to 5.1; 1/2MS+ NAA 0.1 mg/L was the optimizing medium forrooting, and the rooting rate could reach to 100%. The survival rate of young plants was over 99% after theywere transplanted in peat and perlite at the ratio of 2:1. Anomaly plants problem in tissue culture of Chorophytum bichetii could be effectively solved by the control of the cytokinin concentration, and throwing offthe unhealthy plants. The above results would provide scientific basis for micro propagation of Chorophytum bichetii.
