谷氨酰胺对DSS诱导的小鼠结肠炎的治疗作用及机制

【摘要】 目的:探讨谷氨酰胺(Gln)对DSS诱导的小鼠结肠炎的治疗作用及机制。方法:8周龄BALB/c小鼠随机分为3组:Control组:对照;Gln组:DSS诱导结肠炎(4%DSS,7 d)+Gln治疗(1.5 g/kg,4周);DSS组:DSS诱导结肠炎。4周后处死小鼠评估结肠炎症、疾病活动度、上皮细胞凋亡、肠黏膜炎症因子及NF-κb(p65)信号通路。结果:Gln显著降低了DSS模型小鼠的结肠炎症、疾病活动度及上皮细胞凋亡。Gln抑制了肠黏膜NF-κb(p65)信号通路。结论:Gln治疗可减轻肠道炎症,降低肠上皮细胞凋亡,这可能是通过抑制肠黏膜NF-κB(p65)信号通路。

【Abstract】 Objective: To analyse the therapenutic effect of glutamine( Gln) in mice with Ulcerative colitis induced by DSS. Methods: A total of 30 adult mice were randomly divided into Control group,DSS(4% DSS,7 days) and Gln group [DSS treatment and Gln(1. 5 g / kg,4 weeks) ]. Disease activity index( DAI),inflammatory score,TUNEL and Western blot were performed to analyze the effect of gln on colonitis induced by DSS. Results: Gln improved the DAI,inflammatory score and intestinal epithelial cell apoptosis. Western bolt analysis shown Gln depressed the NF-κb( p65) signal path in intestinal mucosa. Conclusion: Gln has significantly therapeutic effect on DSS induced ulcerative colitis through depressing the NF-κB( p65) signal path
