不同浓度促卵泡生成素对小鼠卵母细胞体外成熟的影响

【摘要】 目的探讨不同浓度促卵泡生成素(FSH)对卵母细胞体外成熟(in vitro matueation,IVM)的影响。方法选取8~12周龄雌鼠,腹腔注射HMG,48h后处死取卵巢,穿刺卵巢表面获得卵母细胞。将所得卵母细胞随机分组,置于不同浓度FSH(0,38,75,150,300IU/L)的培养基中,组别由A~E表示。培养24h后,脱去颗粒细胞观察卵子成熟率。对成熟卵行单精子显微注射,观察受精率及胚胎发育情况。结果1C、D、E组较其他组有较高的成熟率,差异有统计学意义(P<0.05),且这三组间差异无统计学意义(P>0.05)。2C、D组较其他组有较高的优质胚胎率(P<0.05),C、D二组间差异无统计学意义(P>0.05),但C组略高于D组。结论培养液中FSH浓度影响IVM结局:1当浓度<75IU/L时,成熟率下降。2当浓度在75~150IU/L时,胚胎发育较好。

【Abstract】 Objective To explore the effects of different concentration of FSH on in vitro maturation of oocytes(in vitro matueation,IVM).Methods Eight to twelve weeks old female rats,48 hours after intraperitoneal injection of HMG,were sacrificed to obtain the ovary,oocyte puncture ovarian surface.The oocytes were random divided into group A,B,C,D and E,under different concentrations of FSH(0,38,75,150,300IU/L)in the culture medium.After cultured for 24 hours,oocyte maturation rate was observed removing granulosa cells.The mature egg for intracytoplasmic sperm injection,the rate of fertilization and embryonic development were observed.Results The mature rate C,D,of E was higher in group C,D,E than other groups,the difference was statistically significant(P<0.05),but the difference among the three groups was not significant(P>0.05).The quality of embryos in group C,D compared with the other group had a higher rate(P<0.05),no significant differences between the two groups in C,D(P>0.05),but the group C was higher than that in group D.Conclusion The FSH concentration in culture solution effects IVM outcome:When the concentration of<75 IU/L,the mature rate is decline;when the concentration of 75-150IU/L in embryonic development is better.
