益肾达络饮对实验性自身免疫性脑脊髓炎小鼠CREB、P-CREB表达的影响

【摘要】 目的观察中药复方益肾达络饮治疗的实验性自身免疫性脑脊髓炎(EAE)小鼠中枢神经系统中c AMP反应元件结合蛋白(CREB)磷酸化水平,探讨其对小鼠中枢神经损伤的可能作用机制。方法建立小鼠EAE动物模型,采用蛋白免疫印迹技术检测正常对照组、模型组、中药组、激素组、中药加激素组、抑制剂组小鼠脑和脊髓中CREB、P-CREB的表达量,并计算CREB磷酸化水平(P-CREB/CREB)。结果在脑组织中,中药组P-CREB相对表达量及CREB磷酸化水平(P-CREB/CREB)与其余各组比较均显著升高(P<0.01);中药组、中药加+激素组和激素组CREB磷酸化水平依次降低(两两比较P<0.01)。在脊髓组织中,中药组P-CREB相对表达量均高于模型组(P<0.01)、对照组(P<0.05)和抑制剂组(P<0.05);中药组、中药+激素组及激素组CREB磷酸化水平均高于模型组(P<0.05),这3组之间两两比较差异无统计学意义(P>0.05)。结论益肾达络饮对EAE的治疗作用可能与升高CREB的磷酸化水平有关。

【Abstract】 Objective To Investigate the phosphorylation level of CREB( c AMP response element-binding protein) in central nervous system of experimental autoimmune encephalomyelitis( EAE) mice,in order to explore the mechanism of Yishendaluo decoction on the therapy of central nervous system injury. Methods EAE mice models were established,and different interference therapies were treated with Yishendaluo decoction-treated,hormone-treated,Yishendaluo decoction-hormone treated and inhibitor-treated groups,every group had four mice. Western blot was used to determine the expressions of CREB and P-CREB proteins in brain and spinal cord tissue of the EAE mice.Results In mice brain tissue,the expression of P-CREB / CREB in Yishendaluo decoction-treated group was higher than that of other groups( all P<0. 01); the phosphorylation level of CREB was declining in Yishendaluo decoction-treated,Yishendaluo decoction-hormone treated and only hormone-treated groups( all P>0. 05). In spinal cord tissue,the level of P-CREB protein in Yishendaluo decoction-treated group was higher than that of model group( P<0. 01),normal group( P<0. 05) and SB203580-treated group( P<0. 05); The phosphorylation levels of CREB in Yishendaluo decoction-treated,Yishendaluo decoction-hormone treated and only hormone-treated groups were higher than those of model group( P<0. 05),but there’s no significant difference among these three groups( all P>0. 05). Conclusions To a certain extent,the mechanism of Yishendaluo decoction on EAE mice might be associated with the phosphorylation level of CREB.
