通心络联合阿托伐他汀、阿司匹林对家兔动脉粥样硬化早期血管外膜炎症因子表达的影响

Effects of Tongxinluo combined with atorvastatin and aspirin on the adventitia inflammation in the early stage of atherosclerosis

【摘要】 目的探讨"金三角"(ATS)方案即通心络联合阿托伐他汀、阿司匹林对家兔动脉粥样硬化早期颈动脉外膜炎症因子表达的影响。方法高脂饮食复合单侧颈总动脉硅胶管包裹制备兔动脉粥样硬化早期模型。72只新西兰兔随机分为对照组、模型组、通心络(TXL)组、阿托伐他汀(ATO)组、阿司匹林(ASP)组和ATS组,每组12只。对照组给予普通饲料、模型组及各用药组家兔均实施单侧颈动脉硅胶管包裹术复合高脂饲料喂养,TXL组给予通心络超微粉混悬液0.3 g·kg-1·d-1灌胃,ATO组给予阿托伐他汀2.5 mg·kg-1·d-1灌胃,ASP组给予阿司匹林12 mg·kg-1·d-1灌胃,ATS组给予Tx L、ATO、ASP三者联合,剂量上,灌胃,连续给药,4 w后取材,HE染色判定颈动脉内中膜病理形态变化,生化法检测血脂变化,ELISA法测定各组家兔颈动脉包裹段外膜单核细胞趋化蛋白(MCP)-1、白介素(IL)-1β、IL-10表达水平,免疫组化法检测血管外膜IL-8表达水平。结果与对照组比较,模型组总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)表达明显增强(P<0.01)。与模型组比较,除ASP组外,各用药组TC、TG、LDL-C表达明显减弱(P<0.01)。与其他用药组比较,ATS组TC、TG、LDL-C表达明显减弱(P<0.01)。与对照组比较,模型组颈动脉外膜中MCP-1、IL-1β表达明显增强,IL-10表达减弱(均P<0.01)。与模型组比较,各用药组颈动脉外膜中MCP-1、IL-1β表达减弱(P<0.01),IL-10表达增强(P<0.05)。与其他用药组比较,ATS组MCP-1、IL-1β表达明显减弱,IL-10表达明显增强(均P<0.01)。各用药组颈动脉外膜IL-8表达减少。结论 TXL、ATO、ASP三药联用可通过降脂、调节血管外膜炎症反应从而有效延缓动脉粥样硬化进程,且较单药应用作用更佳。

【Abstract】 Objective To discuss the effect of Tongxinluo combined with atorvastatin( ATO) and aspirin( ASP) on the adventitia inflammation of early atherosclerosis lesions in rabbits. Methods Seventy-two New Zealand rabbits were randomly divided into control,model,tongxinluo( TXL),ATO,ASP,golden triangle( Tongxinluo combined with ATO and ASP,ATS) groups( n = 12). All the groups were fed with medicine for 4 weeks. The tissue slice of carotid artery was stained by HE and observed under light microscope. The changes of blood liquid were detected by biochemical assay. ELISA was used to measure the carotid artery adventitia of MCP-1,IL-1β,IL-10. The immunohistochemical staining was used to detect the protein expression of IL-8 around the carotid artery adventitia. Results Compared with those of normal group,the contents of TC,TG,LDL-C were significantly increased( P < 0. 01),the content of IL-10 was significantly decreased( P < O. 01) in model group. Compared with those of model group,the contents of TC,TG,LDL-C were significantly decreased( P < 0. 01,P < 0. 05) in TXL and ATO groups,but no significant difference between TXL and ATO groups( P > 0. 05). In ATS group,the contents of TC,TG,LDL-C were lower than those in ATO and TXL groups( P < 0. 01). Compared with those of normal group,the contents of MCP-1,IL-1β were significantly increased( P < 0. 01),the content of IL-10 was significantly decreased( P < 0. 01) in model group. Compared with those of model group,the contents of MCP-1,IL-1βwere decreased( P < 0. 01) in TXL,ATO and ASP groups,but no significant differences between the three groups( P> 0. 05). The content of IL-10 was decreased( P < 0. 01,P < 0. 05) in ATS group,the contents of TC,TG,LDL-C were lower than those in TXL,ATO and ASP groups( P < 0. 01). The content of IL-8 was decreased in TXL,ATO,ASP and ATS groups. Conclusions The ATS project could enhance the effect of regulating lipid metabolism and inhibiting the adventitia inflammation. It played an important role to intervene in the process of atherosclerosis.
