ICF45相互作用蛋白的筛选及鉴定
Screening and identification of proteins interacting with ICF45
【摘要】 为了利用蛋白质组学技术,寻找可能与ICF45存在相互作用的蛋白,从而为探讨ICF45蛋白的生物学功能和作用机制提供依据,本文利用免疫共沉淀和GST-pull down技术对Hela细胞的ICF45蛋白进行富集,通过SDS-PAGE电泳寻找差异条带,并联用质谱进行蛋白鉴定。结果表明:2种实验方法共同筛选得到线粒体定位的分子伴侣HSPA9及Annexin A2与ICF45在体内可能存在相互作用。研究成果为进一步研究ICF45在细胞增殖和调控中的作用奠定了实验基础。
【Abstract】 To identify the potential interacting proteins with ICF45 by proteomics analysis and investigate the biological role and the mechanism of ICF45,the interaction complex of ICF45 was enriched by co-immunoprecipitation and GST-pull down in Hela cells,which was then separated by SDS-PAGE.The candidate proteins were identified by mass spectrometry.The results indicate mitochondrial localization chaperones HSPA9 and Annexin A2 are identified as the possible interaction protein with ICF45 by two methods,respectively.The function of ICF45 and potential interacting proteins will provide experimental basis for identifying the role of ICF45 on cell proliferation and the regulation.
