力达霉素抑制肝癌细胞球形成的研究

【摘要】 目的本文运用肝癌细胞球模型,比较力达霉素与常用化疗药物对肝癌细胞球的作用效果,并从调节耐药蛋白角度探讨力达霉素作用机制。方法建立肝癌Huh7细胞球1-3代培养传代方法,检测其肿瘤干细胞特性;用IC50浓度的不同化疗药物作用后,观察肿瘤细胞球大小和计数;检测不同药物作用后ABCG2 m RNA和蛋白的变化。结果肝癌细胞球1~3代中,能稳定的表达肿瘤干细胞的特性;力达霉素能显著抑制1~3代肿瘤细胞球的形成,效果优于多柔比星和顺铂,力达霉素能显著降低肝癌细胞球中的ABCG2基因和蛋白的表达。结论肝癌Huh7细胞球具有肿瘤干细胞特性,能有效的筛选出具有抑制肿瘤干细胞的药物;力达霉素能抑制肝癌细胞球的形成可能与其降低ABCG2表达逆转耐药有关。

【Abstract】 Objective To compare the effect of lidamycin and chemical drugs on hepatic cancer spheres, and investigate the mechanism of lidamycin on drug-resistant protein. Methods Spheres of Huh7 cells in hepatocellular carcinoma were cultured and passaged from 13 generations. Characteristics of cancer stem cells were measured. The size and number of spheres were observed after treatment of IC50 concentration of different drugs. Levels of ABCG2 m RNA and protein were tested by different drugs. Results In 13 generations, Characteristics of cancer stem cells were stably expressed. The formations of spheres were significantly reduced by lidamycin during 13 passages, better than DOX and CDDP. Expression of ABCG2 m RNA and protein were decreased by lidamycin. Conclusion This study indicates that the spheres model in HCC is partially represented the characteristics of cancer stem cells and effective for the drug screening. It suggests that mechanism of lidamycin on spheres might be connected with modulation of ABCG2 expression to reverse drug resistance.
