不同力学刺激对骨性关节炎软骨Ⅰ型胶原蛋白产生的影响

【摘要】 [目的]观察异常力学刺激对骨性关节炎病程中软骨I型胶原蛋白表达的影响,探讨骨性关节炎软骨中I型胶原蛋白的异常表达机理。[方法]18只SD大鼠,将动物随机分为A(力学刺激组)、B(对照组)、C(假手术组)3组,每组6只。A、B组按照标准方法行前交叉韧带横断术造骨性关节模型,C组作为假手术组。术后1周将A组与C组置于动物跑台上给予力学刺激:8米/min,30 min,5次/周,B组养在鼠笼中正常饮食、喝水、活动。术后5周处死所有动物,取样行HE染色和番红O-快绿染色,免疫组化法检测I型胶原在各组关节软骨中的表达;根据改良Mankin评分对标本进行病理分期;IPP软件分析各组I型胶原蛋白表达情况,研究异常力学刺激与I型胶原蛋白表达的相关性。[结果]A组膝关节软骨I型胶原蛋白的表达高于番红O-快绿染色,示A组软骨退化程度最高,B组次之,C组最低;Mankin评分A组高于B、C两组(P<0.05),C组最低。[结论]异常的力学刺激与骨性关节炎病程中I型胶原蛋白的产生具有相关性,可能是导致骨性关节炎软骨中I型胶原蛋白产生的原因之一。

【Abstract】 [Objective] The purpose of this study was to investigate the role of mechanical stimulations in regulating the expression of type I collagen in chondrocyte to explore the mechanism of expression of type I collagen in the course of OA.[Methods] Eigthteen Sprague Dawley rats were divided into three groups,srength mechanical stimulation group( A),control group( B),Sham group( C),group A and B were subjected to anterior cruciate ligament transaction( ACLT). Running was performed on a motorized treadmill after 1 week,at a speed of 8m / min( group A and group C) for 30 min / day for 5 days per week. Five weeks later,OA histopathology in the knees was evaluated using the HE and safranin O- fast green staining,immunohistochemistry( COL- I),the Mankin score were analyzed. [Results] Levels of collagen type I highly expressed and the Mankin score deteriorated in the group A after 5 weeks and the ratio suggested the development of OA. The level of collagen type I was lower,and Mankin score was also lower in group B. The level of COL- I expression in the group C was apparently lower than that in other two groups,and Mankin score was lowest. [Conclusion] The level of COL- I expression in cartilage is positively correlated with mechanic stimulation.
