急性百草枯中毒大鼠肺损伤与水通道蛋白1和5表达的相关性研究
To correlation between aquaporin 1,5 expression and lung injuries in acute paraquat poisoning rats
【摘要】 目的 观察水通道蛋白1、5(AQP1、AQP5)表达与急性百草枯中毒大鼠肺损伤(ALI)的相关性,并探讨其作用机制。方法将实验大鼠分为百草枯中毒组(按第1、3、5天分亚组)及生理盐水对照组;中毒组(每亚组8只)腹腔注射百草枯,对照组(5只)在相同时间点给予等量的生理盐水腹腔注射;对比两组大鼠的肺组织病理学改变、肺湿重/干重比(W/D)及AQP1、AQP5的表达。结果光镜下见,百草枯中毒肺组织第1天与对照组比较,无明显改变;第3天肺毛细血管扩张、充血,肺泡腔内渗出、出血、肺泡呈代偿性肺气肿改变;第5天肺泡上皮细胞增生、肥大,肺泡腔及肺间质内中性粒细胞渗出,局部形成脓肿。肺W/D第1天与对照组比较差异无统计学意义;第3、5天明显高于对照组(P<0.01);第1、5天与第3天亚组比较差异有统计学意义(P<0.01)。中毒组血清及肺组织匀浆AQP1、AQP5定量与对照组比较差异有统计学意义(P<0.01);其中中毒第3天升高最明显;第1、3、5天组间比较差异有统计学意义(P<0.01)。血清AQP1、肺组织匀浆AQP1及血清AQP5定量第1、3天均呈量效关系。结论 AQP1、AQP5参与百
【Abstract】 Objective To study the relevance between expressions of aquaporin 1,5(AQP1,AQP5) and lung injury induced by acute paraquat poisoning in rats,and the mechanism involved.Methods Randomly divided into two groups of paraquat poisoning(including 1,3,5 days three subgroups) and normal saline control group,experimental sub group of eight,control group five;rats in paraquat poisoning group were treated by intraperitoneal injection of paraquat,while those in control group were injected physiological saline instead;histopathology of animal pathology,lung wet/dry weight ratio,expression of AQP 1,5 in rats of both groups were observed and compared.Results No difference of lung tissue of paraquat poisoning group and control group had been observed under light microscope in 1 day subgroup.Pulmonary capillary congestion,alveolar infiltration and hemorrhage,compensatory emphys ema alveolar were observed in 3 days sub- group.Alveolar epithelial hyperplasia and hypertrophy,alveolar and interstitial lung neutrophil infiltration,local abscess formation emerged in5 days subgroup.Lung wet/dry weight ratio of control group had no difference with 1 day subgroup,but significantly lower than 3,5 subgroups(P < 0.01).Differences among 1,3,5 days subgroups also reached significant level(P<0.01).Quantitative of AQP1,5 in serum and lung homogenate results showed that the highest level of it was in 3 days subgroup while 1,3,5 days subgroups significantly differed to each other.At the same time,difference between poisoning group and control group were significant(P<0.01).Moreover,quantity of serum AQP1,AQP1 in lung homogenate,and serum AQP5 in 1,3 day sub groups showed a dose effect relationship with paraquat used.Conclusion AQP1,AQP5 are involved in the formation of acute lung injury in paraquat poisoning.The expression of AQP1,5 is related with the degree of pulmonary edema.
