负载阿霉素的透明质酸修饰还原敏感纳米粒子对乳腺癌细胞株MDA-MB231的靶向杀伤及生物学意义

【摘要】 目的研究负载阿霉素(doxorubicin,DOX)的透明质酸修饰还原敏感纳米粒子HA-Lys-LA对乳腺癌细胞株MDA-MB231的靶向性及其抗肿瘤效果。方法通过流式细胞术检测人乳腺癌细胞株MDA-MB231细胞表面CD44分子的表达,检测对负载DOX的透明质酸修饰还原敏感纳米粒子HA-Lys-LA(DOX-NPs)的摄取情况;利用CCK-8试剂盒检测DOX-NPs对乳腺癌细胞株MDA-MB231杀伤情况;通过划痕实验研究DOX-NPs对乳腺癌细胞株MDA-MB231细胞迁移的抑制情况;利用Hoechst染色法检测DOX-NPs对MDA-MB231细胞凋亡的诱导情况。结果 DOX-NPs能够被高表达CD44分子的人乳腺癌细胞株MDA-MB231细胞有效摄取;DOX-NPs对MDA-MB231细胞的增殖、迁移具有显著抑制作用,并明显诱导其凋亡。结论负载DOX的透明质酸修饰还原敏感纳米粒子DOX-NPs对人乳腺癌细胞株MDA-MB231细胞具有靶向杀伤及诱导凋亡作用。
【Abstract】 Objective To investigate the effect and significance of doxorubicin loaded reversibly crosslinked hyaluronic acid nanoparticles(DOX-NPs) on targeting and inhibiting breast cancer cell MDA-MB231. Methods The expression of CD44 on human breast cancer cell line MDA-MB231 and the uptake of DOX-NPs by MDA-MB231 were measured by flow cytomery assay. The proliferation, apoptosis and migration of MDA-MB231 treated with DOX-NPs were determined by CCK-8assay, Hoechst staining and wound healing experiment, respectively. Results DOX-NPs could be uptaked by highly expressed CD44 of MDA-MB231 cells. DOX-NPs had obvious inhibitional effect on the cells proliferation and migration of MDAMB231 cells, it could also induce apoptosis. Conclusion DOX-NPs can actively kill the breast cancer cells MDA-MB231 targetly, suppress the proliferation and induce apoptosis.
