芙蓉散质量标准的建立
Establishment of Quality Standards for Lotus Powder
【摘要】 目的:建立院内制剂芙蓉散的质量评价标准。方法:采用薄层色谱法对芙蓉散中黄芩、大黄、小檗碱和冰片进行定性鉴别;采用高效液相色谱法(HPLC)方法,色谱柱:Kromasil C18(150 mm×4.6 mm,5μm),流动相:甲醇-水-磷酸(47∶53∶0.2);流速:1 m L/min;检测波长:280 nm;柱温:35℃;测定芙蓉散中黄芩苷的含量。结果:黄芩、大黄、小檗碱和冰片薄层色谱法(TLC)鉴别具有较好的专属性,阴性对照无干扰。以黄芩苷进样浓度(X)对峰面积(Y)进行线性拟合,得黄芩苷回归方程为:Y=5.494 06e-007X-0.003 888 11,r=0.9997;线性范围为1.242~0.0828μg呈现良好线性关系。测得黄芩苷平均回收率为98.3%,RSD为1.94%。结论:该方法简便、准确、专属性及重现性好,可作为芙蓉散的质量控制方法。
【Abstract】 ObjectiveTo establish the quality evaluation standard of hospital preparation of lotus powder.MethodsFor the identification of Radix Scutellariae,rhubarb,lotus powder berberine and borneol TLC andHPLC methods were used,the conditition of column chromatography was Kromasil C18(150 mm×4.6 mm,5 μm),the mobile phase of methanol-water-phosphate(47 ∶ 53 ∶ 0.2);the velocity:1 m L/min,and the detection wavelength:280 nm;the column temperature:35℃;and determination of the content of Baicalin in lotus powder wasdone.ResultsBaicalin,rhubarb,berberine and borneol TLC identification showed good specificity,negativecontrol without interference,the baicalin in sample concentration(x) of the linear fit to the peak area(y),baicalinregression equation showed Y=5.49406e-007X-0.00388811,r=0.9997;linear range for 1.242 μ g~0.0828 μ gshowed good linear relationship.The average recovery of baicalin was found 98.3%,and RSD 1.94%.ConclusionThis method is simple,accurate,specific and reproducible,so can be used for quality control oflotus powder.
