平菇固体发酵菌丝体次级代谢产物的研究
Study on secondary metabolites from the solid culture of Pleurotus ostreatus
【摘要】 目的研究平菇菌丝大米固态发酵的次级代谢产物,以期从中找到食药用真菌预防保健的物质基础。方法采用正相硅胶柱色谱、ODS柱色谱、Sephadex LH-20、高效液相色谱等方法进行分离纯化;利用理化性质和波谱分析对单体化合物进行结构鉴定。结果从平菇菌丝大米固态发酵物中分离得到了13个化合物,分别为:甘露醇(1)、(22E,24R)-24-甲基-麦角甾-7,22-二烯-3β,5α,6α-三醇(2)、5,7-二甲氧基苯酞(3)、4,4’-氧二丁酸(4)、对羟基苯甲醛(5)、苯甲酸(6)、吲哚-2,3-二酮(7)、对甲氧基苯甲酸(8)、反式桂皮酸(9)、3-吲哚甲醛(10)、(R)-3-羟基-3-(2’-氧代丙基)吲哚-2-酮(11)、(22E,24R)-3β,5α,9α-三羟基-麦角甾-7,22-二烯-6-酮(12)、尿嘧啶核苷(13)。结论其中化合物4、8、11、13为首次从侧耳属中分离得到,化合物3、7、9、10、12为首次从平菇中分离得到,化合物4为新天然产物。
【Abstract】 The ethyl acetate extracts of the solid cultrue of Pleurotus ostreatus were subjected to silica gel ODS,sephadex LH-20 column chromatography and HPLC prepration. Thirteen compounds were isolated from the solid culture,and their structures were identified as mannitol( 1),( 22 E,24R)-24-methyl-ergosta-7,22-diene-3β,5α,6α-triol( 2),5,7-dimethoxyisobenzofuran-1( 3H)-one( 3),4,4’-oxydibutyric acid( 4),p-hydroxybenzaldehyde( 5),benzoic acid( 6),indole-2,3-dione( 7),4-methoxybenzoic acid( 8),trans-cinnamic acid( 9),1H-indole-3-carboxaldehyde( 10),( R)-3-hydroxy-3-( 2’-oxopropyl) indolin-2-one( 11),( 22 E,24R)-3β,5α,9α-trihydroxy-ergosta-7,22-dien-6-one( 12),uridine( 13),on the basis their spectroscopic and physico-chemical properties. Among them,compounds 4,8,11 and 13 have been obtained from Pleurotus genus for the first time and compounds 3,7,9,10,12 were obtained from the species firstly. Meanwhile,compound 4 is a newnatural product.
