蜂毒制剂“神蜂精”对佐剂性关节炎大鼠血管表皮生长因子及其受体的影响

【摘要】 目的:探讨"神蜂精"对佐剂性关节炎大鼠的治疗的可能机制。方法:60只SD雄性大鼠进行随机性分组,分为正常组、模型组、"神蜂精"高剂量组(1.0m L/kg/d)、"神蜂精"中剂量组(0.5m L/kg/d)、"神蜂精"低剂量组(0.25m L/kg/d)、阳性组(复方醋酸地塞米松乳膏给药组,1.0g/kg/d),每组10只。除正常组外,其余组大鼠右后足跖皮下注射完全弗氏佐剂诱导产生佐剂性关节炎(AA)。造模14 d后连续给药28 d。测量注射足足肿胀体积,采用酶联免疫吸附法(ELISA法)测定大鼠血清中血管表皮生长因子(VEGF)、血管表皮生长因子受体-1(VEGFR-1)及血管表皮生长因子受体-2(VEGFR-2)的含量。结果:(1)与模型组相比,"神蜂精"治疗组均能显著降低大鼠注射足足爪肿胀度(p<0.01);(2)与模型组相比,"神蜂精"高剂量组、中剂量组大鼠血清中VEGF和VEGFR-1含量,极显著降低(p<0.01,p<0.05);(3)各组大鼠血清中VEGFR-2的含量无统计学差异(p>0.05)。结论:"神蜂精"明显的抑制佐剂性关节炎大鼠足部炎症,降低血清VEGF及VEGFR-1的表达,从而减轻关节病理性血管新生。

【Abstract】 Objective:To investigate the mechanism of Shen Fengjing(SFJ) for adjuvant arthritic(AA)rats.Methods:Complete Freund’s adjuvant(CFA) was injected to induce experimental arthritis in rats. 60 SD male rats were randomly divided into the normal group, model group, SFJ high-dose group, SFJ middle-dose group, SFJ low-dose group,Compound Dexamethaso(CD)group with 10 rats in each. A model of experimental adjuvant-induced arthritic rats(AA rats) was established in the latter five groups. After 14 days, the six groups were treated with normal saline, 1.0m L/kg SFJ and 0.5, 0.25 m L/kg SFJ, 1.0g/kg CD by local treatment for 28 days. Paw swelling degree of AA rats was measured.The content of vascular endothelial growth factor, vascular endothelial growth factor receptor-1 and vascular endothelial growth factor receptor-2(VEGF, VEGFR-1, VEGFR-2) in serum were measured by Enzyme-linked Immunosorbent Assay(ELISA) kits. Results:(1) Compare with the model group, all SFJ-treated groups significantly decreased the paw swelling degree(p<0.01);(2) Compare with the model group, High SFJ-treated group and middle SFJ-treated group significantly decreased the content of VEGF and VEGFR-1 in the serum of rats(p<0.01, p<0.05);(3) Compare with the model group, the content of VEGFR-2 in the serum of all groups had no significant difference(p>0.05). Conclusions: SFJ could significantly inhibit the inflammation of the AA rats, and decreased the content of VEGF and VEGFR-1 of AA rats, then reduced joint pathological?angiogenesis.
