双氢睾酮对三阴乳腺癌MDA-MB-231细胞增殖的抑制作用

【摘要】 目的:研究不同浓度双氢睾酮(DHT)对三阴乳腺癌MDA-MB-231细胞增殖的抑制作用。方法:采用2-(2-甲氧基-4-硝苯基)-3-(4-硝苯基)-5-(2,4-二磺基苯)-2H-四氮唑单钠盐(CCK8)比色法检测1、10、100 nmol/L的DHT分别作用24、48、72 h后对MDA-MB-231细胞增殖的影响;采用流式细胞检测仪分别检测上述浓度DHT作用48 h后对MDA-MB-231细胞周期的影响;并用Agilent mi RNA microarray芯片V19杂交技术分析100 nmol/L DHT作用48 h前后细胞的mi RNA表达谱,以生物学软件SAS筛选出表达差异在5倍以上的mi RNA。结果:1、10、100 nmol/L的DHT对MDA-MB-231细胞增殖均有抑制作用,且与DHT浓度和作用时间呈正相关。与作用前比较,DHT作用于细胞48 h后S期细胞增多,出现S期阻滞,且与DHT浓度呈正相关。100 nmol/L DHT作用于细胞48 h后较作用前有9个mi RNA表达上调大于5倍,10个mi RNA表达下调大于5倍。结论:DHT对MDA-MB-231细胞增殖具有较为明显的抑制作用,其机制可能与阻滞细胞周期及诱导细胞mi RNA表达谱改变有关。
【Abstract】 OBJECTIVE:To study the inhibitory effects of dihydrotestosterone(DHT)with different concentrations on proliferation of triple negative breast cancer cells MDA-MB-231. METHODS:2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfoanilino)-2H-tetrazolium sodium salt(CCK-8)was used to determine the effects of drugs with three concentrations(1,10,100nmol/L) on cells proliferation after 24 h,48 h and 72 h. The effect of DHT with different concentrations on the cycle of MDA-MB-231 was detected by flow cytometry after 48 h. The expression of mi RNA was analyzed by the Gilent mi RNA microarray chip V19 hybridization before and after 48 h of 100 nmol/L DHT treatment,and the differential expression of mi RNA over 5times was screened by bioinformatics software SAS. RESULTS:DHT of 1,10 and 100 nmol/L could inhibited MDA-MB-231 proliferation,and had positive correlation with concentration and action time. Compared with before treatment,MDA-MB-231 cells in S phase increased in a dose-dependent way after 48 h of DHT administration and had positive correlation with concentration. Compared with before treatment,there were 9 mi RNAs up-regulated and 10 down-regulated over 5 times in cells with DHT administration after 48 h. CONCLUSIONS:The inhibitory effect of DHT on proliferation of MDA-MB-231 cells was obvious and the mechanism might be correlated to de-regulating cellular cycles and changing mi RNA expressions.
