酸枣仁汤对睡眠剥夺模型大鼠学习记忆的影响

【摘要】 目的:研究酸枣仁汤对睡眠剥夺模型大鼠学习记忆的影响。方法:采用改良多平台法复制大鼠睡眠剥夺模型。50只SD大鼠随机均分为正常对照(等容蒸馏水)组、模型(等容蒸馏水)组与酸枣仁汤高、中、低剂量[21.00、10.50、5.25 g(生药)/kg]组,复制模型的同时ig给药,每天1次,连续7 d。观察大鼠一般状态,测定体质量;Y型迷宫测定大鼠错误反应次数与主动回避率;检测大鼠海马组织乙酰胆碱酯酶(ACh E)、乙酰胆碱转移酶(Ch AT)活性与血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。结果:与正常对照组比较,模型组大鼠一般状态欠佳,体质量减少;错误反应次数增加,主动回避率降低;海马组织ACh E活性增强、Ch AT活性减弱,血清SOD活性减弱、MDA含量增加,差异均有统计学意义(P<0.01或P<0.05)。与模型组比较,酸枣仁汤高剂量组大鼠体质量增加;错误反应次数减少,主动回避率增加;高、中剂量组大鼠海马组织ACh E活性减弱、Ch AT活性增强,血清SOD活性增强;高、中、低剂量组大鼠血清MDA含量减少,差异均有统计学意义(P<0.01或P<0.05)。结论:酸枣仁汤能改善睡眠剥夺模型大鼠的学习记忆下降,其机制可能与保护胆碱能系统和抗氧化作用有关。

【Abstract】 OBJECTIVE:To study the effects of Ziziphus jujuba decoction on the learning and memory in sleep deprived rats.METHODS:The model rats of sleep deprivation were established with modified multiple platform method.50 SD rats were randomly divided into normal control(isovolumic distilled water)group,model(isovolumic distilled water)group and Z.jujuba decoction high-dose,medium-dose and low-dose [21.00,10.50,5.25 g(medicinal materials)/kg] groups.Replication model was conducted with intragastric administration for 7 d,once a day.General conditions of rats were observed,and body mass was weighed.The error response times and active avoidance rate of rats were tested by Y-maze.The activity of Ch AT and ACh E in rat hippocampal,SOD activity and MDA content in rat serum were determined.RESULTS:Compared with normal control group,rats had poor general conditions,the weight decreased,the error response times increased,the active avoidance rate decreased,ACh E activity increased,Ch AT activity in hippocampal and SOD activity in serum decreased,and MDA content in serum increased in model group.There was statistical significant difference(P<0.01 or P<0.05).Compared with model group,weight of rats increased,active avoidance rate and the error response times decreased in Z.jujuba decoction high-dose group.ACh E activity in hippocampal decreased,Ch AT activity in hippocampal and SOD activity in serum increased in Z.jujuba decoction high-dose group and mediumdose group.the MDA content in serum decreased in Z.jujuba decoction high-dose group,medium-dose and low-dose groups.There was statistical significant difference(P<0.01 or P<0.05).CONCLUSIONS:Z.jujuba decoction can improve the learning and memory in model rats after sleep deprivation.It is possibly related to the protection of cholinergic system and its antioxidant effect.
