星点设计-响应面法优化维药刺山柑果生物碱提取工艺
Optimization of Extraction Technology for Total Alkaloids from Uygur Medicine Capparis spinosa Fruit by Central Composite Design-Response Surface Methodology
【摘要】 目的:优化维吾尔药刺山柑果总生物碱的提取工艺。方法:以乙醇体积分数、加醇倍量、提取时间为自变量,以刺山柑果中总生物碱(以甜菜碱为对照)含量为因变量,应用Design-Exepert 8.0.5软件,通过对自变量各水平的多元线性回归及二项式拟合,用响应面法优化出最佳提取工艺,同时进行预测值与实测值比较分析。结果:二项式拟合复相关系数R2=0.955 8,回归方程代表性较好;确定最佳提取工艺为10倍量90%的乙醇提取2次,每次1.5 h;经5次验证试验,最佳工艺提取的刺山柑果总生物碱含量为77.99 mg/g(RSD=0.95%,n=5),与预测值78.33 mg/g比较偏离率小于2%。结论:星点设计-响应面法优化刺山柑果总生物碱的提取工艺方法简单,精密度高,模型可靠,可预测性趋势良好。
【Abstract】 OBJECTIVE:To optimize the extraction process of total alkaloids from Uygur medicine Capparis spinosa fruits.METHODS:The ethanol concentration and amount,extraction time as independent variables,and the content of total alkaloids from C. Spinosa fruits(oxyneurine as control)as dependent variable were regressed multiple linearity and fitted by binomial expression using Design-Exepert 8.0.5 statistical software. Response surface methodology was used to optimize the extraction process.Meanwhile,predicted and measured values were compared. RESULTS:The correlation coefficient R2 fitted by binomial expression was equal to 0.955 8,and regression equation was good in representativeness. The optimum extraction progress was as follows:10-fold 90% ethanol,extracting for 2 times,lasting for 1.5 h each time. After 5 times of validation tests,the content of total alkaloids from C. spinosa fruits extracted by optimal extraction technology was 77.99 mg/g(RSD=0.95%,n=5),and bias ratio was lower than 2%,compared with predicted value of 78.33 mg/g. CONCLUSIONS:The extraction technology of total alkaloids from C. spinosa fruits optimized by central composite design-response surface methodology is simple and reliable with high precision and good predictability.
