一株血鹦鹉嗜水气单胞菌的分离鉴定

【摘要】 为确诊天津市某水产养殖场血鹦鹉突发传染病病原,并对其进行有效的预防和治疗,本实验对患病血鹦鹉进行了病原的分离鉴定及生物学特性研究。从病灶部位共分离出一株疑似病原菌,经人工感染后发现编号为YW01的分离株具有较高的致病性,临床症状与自然发病症状一致,LD50为9.55×109cfu/m L。YW01革兰氏染色呈阴性,杆状。16S r RNA基因序列分析结果显示,YW01菌株在系统发育树上与嗜水气单胞菌(Aeromonas hydrophila)聚类,同源性达到99%。综合生理生化特征、形态学观察、人工感染以及16S r RNA基因序列分析,表明YW01分离株为嗜水气单胞菌。YW01分离株的最适生长p H为7~8,最适生长温度为32℃。在32℃200 r/min条件下的生长周期是:0~4 h为生长延迟期,4 h~10 h为对数生长期,10 h~22 h为稳定期,22 h~48 h为衰亡期。药敏试验表明,YW01分离株对多粘霉素B、阿米卡星、诺氟沙星等高度敏感,对磺胺异恶唑中度敏感,对麦迪霉素、氨苄西林、林可霉素、头孢拉定等不敏感。本研究为该病的防治提供了实验依据。

【Abstract】 In order to identify the pathogen which caused a serious disease in a Cichlasoma var. farm of Zhongmin in Ninghe district of Tianjin during June to July 2014, a pathogenic bacteria was isolated from the affected Cichlasoma var. The negative Gram staining bacteria isolate was a high pathogenicity with 9.55 ×109LD50to the birds, which produced the similar clinical symptoms in artificially infected Cichlasoma var. as the naturally infected cases. Eventually, the bacteria isolate was idantified as Aeromonas hydrophila by Gram-stained, VITEK 2 compact automated microbiology system, artificial infection and 16 S r RNA gene analysis. Moreover, the antibiotic test showed that the isolate was sensitive to Polymixin B, Amikacin, Norfloxacin and so on,intermediate sensitivity to Sulfafurazole, and resistance to Midecamycin, Ampicillin, Cillimycin and Cefradine.
