人参皂苷Rg1对大鼠急性心肌缺血抗氧化损伤指标及超微结构的影响

【摘要】 目的 :研究人参皂苷Rg1对大鼠急性心肌缺血抗氧化损伤指标及超微结构的影响。方法 :建立大鼠急性心肌缺血模型,大鼠随机分为假手术组、急性心肌缺血模型组(模型组)、人参皂苷Rg1 5 mg/kg组和人参皂苷Rg1 10 mg/kg组,腹腔注射给药两周后取材。氯化三苯基四氮唑(TTC)法测量心肌梗死面积,电子显微镜观察心肌组织超微结构变化,比色法测定大鼠血清超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活性,以及丙二醛(MDA)含量。结果 :与模型组比较,人参皂苷Rg1 5mg/kg组和人参皂苷Rg1 10 mg/kg组可明显减少心肌梗死面积,减少心肌组织损伤,人参皂苷Rg1 10 mg/kg可明显提高大鼠血清SOD和GSH-Px活性,降低丙二醛的含量。结论 :人参皂苷Rg1对急性心肌缺血具有保护作用,抗氧化损伤作用是其保护作用机制之一。

【Abstract】 Objective: To study the effect of ginsenosides-Rg1 on acute myocardial ischemic anti-oxidative injury indicators and ultra-structure in experimental rats.Methods: The acute ischemia model was established in experimental rats and the animals were randomly divided into 4 groups: Sham operation group, Model group, Ginsenoside-Rg1 5 mg/kg group and Ginsenoside-Rg1 10 mg/kg group. The rats were treated by intraperitoneal injection for 2 weeks. The area of myocardial infarction was measured by 2, 3, 5-triphenyltetrazolium chloride(TTC) staining, the structural change of myocardial tissue was observed by electron microscope. The activities of blood superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and malondialdehyde(MDA) content were examined by spectrophotometry.Results: Compared with Model group, Ginsenoside-Rg1 5 mg/kg group and Ginsenoside-Rg1 10 mg/kg group showed obviously decreased area of myocardial infarction and myocardial tissue damage. Ginsenoside-Rg1 10 mg/kg group presented significantly increased activities of SOD, GSH-Px and decreased content of MDA in the blood. Conclusion: Ginsenoside-Rg1 has the protective effect on acute myocardial ischemia in experimental rats, the antioxidative injury is one of the mechanisms.
