HPLC法分析腐乳氨基酸的条件优化
Optimization of Analysis of Amino Acid in Fermented Bean Curd by HPLC Method
【摘要】 氨基酸的种类及含量对腐乳后发酵过程的研究具有重要的理论和现实意义,所以必须建立一种便捷可靠的分析方法。高效液相色谱分析水平高、检测范围大,与衍生化技术联用后可以增加腐乳样品中氨基酸检测的灵敏度和精确度。对高效液相色谱法测定腐乳中氨基酸的方法进行了色谱条件优化,将其中对色谱峰分离及峰形有重要影响的梯度洗脱条件、柱温、流速进行优化,研究中以邻苯二甲醛和氯甲酸芴甲酯为衍生化试剂,优化后得到的色谱条件为:流动相A:磷酸氢二钠溶液(pH 7.8),流动相B:甲醇-乙腈-水溶液(体积比45∶45∶10),流速为0.6mL/min,温度为50℃,检测波长为338,262nm,进样体积为20μL,精确度及重现性中峰面积相对标准偏差RSD均小于0.2%,回收率在90.4%~118.7%,最终检测出腐乳后发酵过程中共有16种氨基酸。
【Abstract】 The kinds and content of amino acids have important theoretical and practical significance on the research of the post-fermentation of fermented bean curd,so it is necessary to establish a convenient and reliable analysis method.High performance liquid chromatography(HPLC)has high level of analysis and wide detection range,and can increase the sensitivity and accuracy of fermented bean curd samples after combination of derivatization technology.In this paper,the high performance liquid chromatography(HPLC)method is developed for the determination of amino acids in fermented bean curd to optimize the chromatographic conditions, which have important effects on the chromatographic peak separation and shape of gradient elution conditions,column temperature and flow rate,take formaldehyde and benzene chloride acid methyl fluorene as the derivatization reagent,the optimized chromatographic conditions are as follows: mobile phase A:disodium hydrogen phosphate solution(pH 7.8),mobile phase B:methanol,acetonitrile-water solution(volume ratio of45∶45∶10),the flow rate of 0.6mL/min,the temperature of 50 ℃,detection wavelength of 338,262 nm,the sampling volume of 20μL,the relative standard deviation RSD of accuracy and reproducibility is less than 0.2%,the recovery rate of 90.4 %~118.7%,finally 16 kinds of amino acids are detected in the post-fermentation process of fermented bean curd.
